Brucella vaccination is one of the most important strategies for controlling brucellosis in livestock. The A19 strain was the effective vaccine used to control brucellosis in China. Virulence test showed that the A19 was significantly attenuated at chronic infection stage in infected mouse model. In growth analysis, the A19 exhibited a quick growth at exponential phase and premature at stationary phase. The inflammatory response of macrophages infected by the A19 was detected using TaqMan qPCR assay, indicating that the inflammatory level of the A19-infected macrophages was higher than that of the S2308 infection. Cell death analysis showed that the A19 was not cytotoxic for macrophages. Cell infection showed that the A19 reduced its ability to invade, survive and traffic within host cells, and the intracellular A19 hardly excludes lysosome-associated marker LAMP-1, suggesting that the A19 can't escape the lysosome degradation within host cells. In further study, the sensitivity test exhibited that the A19 is more sensitive to stress and bactericidal factors than the S2308 strain, Western blot and silver staining analysis exhibited that the A19 has a different expression pattern of OMPs and reduces LPS O-antigen expression relative to the S2308 strain. Those data give us a more detailed understanding about the A19 vaccine strain, which will be beneficial for improvement of current Brucella vaccine and overcoming its defects.The pathogen concentration in human excreta needs to be managed appropriately, but a predictive approach has yet to be implemented due to a lack of kinetics models for pathogen inactivation that are available under varied environmental conditions. Our goals were to develop inactivation kinetics models of microorganisms applicable under varied environmental conditions of excreta matrices and to identify the appropriate indicators that can be monitored during disinfection processes. We conducted a systematic review targeting previous studies that presented time-course decay of a microorganism and environmental conditions of matrices. Defined as a function of measurable factors including treatment time, pH, temperature, ammonia concentration and moisture content, the kinetic model parameters were statistically estimated using hierarchical Bayesian modeling. The inactivation kinetics models were constructed for Escherichia coli, Salmonella, Enterococcus, Ascaris eggs, bacteriophage MS2, enterobacteria phage phiX174 and adenovirus. The inactivation rates of a microorganism were predicted using the established model. Ascaris eggs were identified as the most tolerant microorganisms, followed by bacteriophage MS2 and Enterococcus. Ammonia concentration, temperature and moisture content were the critical factors for the Ascaris inactivation. Our model predictions coincided with the current WHO guidelines. The developed inactivation kinetics models enable us to predict microbial concentration in excreta matrices under varied environmental conditions, which is essential for microbiological risk management in emerging resource recovery practices from human excreta.Introducing wild crops and plants complements intentions for improved biodiversity outcomes in agricultural landscapes; and provides opportunity for in-situ conservation of a diverse range of wild plants and crops, and improved connectivity between conserved areas. This opinion article considers definitions and common value of conserving wild crops and plants, and crop wild relatives, in-situ and ex-situ. Rewilding definitions and common uses for policy and practice are described. The Ecological Sensitivity within Human Realities (ESHR) concept was developed to guide human natural-environment interactions in agricultural systems and landscapes for improved functional biodiversity outcomes. It is used to assess possible appropriateness of rewilding for agricultural systems and landscapes. The assessment demonstrates how agricultural systems and landscapes are often excluded, despite appearing a useful term to encourage such conservation efforts. The importance of a more specific term rather than a more specific definition and use of rewilding is suggested. Agricultural wilding is introduced as a more specific term for introducing and conserving wild crops and plants for agricultural purposes, as wild productive systems, useful for policy or other approaches the guide human natural-environment interactions. The conceptual approach to the article provides theoretical suggestions for minimum proportions of wild crops and plants for wild productive systems according to native and non native landscapes. From a conceptual to applied discussion, relevance of agricultural wilding for coffee farming landscapes is explained in some depth, and for an existing biodiversity campaign and an EU agricultural policy, briefly. MicroRNAs (miRNAs) have engaged in the progression of myocardial infarction (MI). Nevertheless, the mechanism of miR-150-5p in MI is still in its infancy. Therefore, the present study was set out to investigate the effect of bone marrow mesenchymal stem cells derived exosomes (BMSCs-Exo) and miR-150-5p in MI via regulating B-cell lymphoma-associatedX(Bax). BMSCs-Exo were isolated and extracted, and exosomes with miR-150-5p agomir or antagomir was constructed. Then, a mouse MI model was induced by ligation of left anterior descending coronary artery. Mice were injected with exosomes and miR-150-5p agomir/antagomir to detect cardiac function, pathological changes, and apoptosis rate of cardiomyocytes. miR-150-5p and Bax expression in myocardial tissues were tested. The targeting relationship between miR-150-5p and Bax was verified. BMSCs and exosomes were successfully extracted. BMSCs-derived exosomal miR-150-5p improved cardiac function, alleviated pathological changes of myocardium, decreased apoptosis rate of cardiomyocytes in MI mice. miR-150-5p expression was reduced and Bax expression was elevated in myocardial tissues of MI mice, while exosomes raised miR-150-5p expression and reduced Bax expression in MI mice. miR-150-5p was found to target Bax. On all accounts, the present study provides evidence that BMSCs-derived exosomal miR-150-5p attenuates apoptosis of cardiomyocytes and improves cardiac function of MI mice via targeting Bax.
