However, disease progression or early relapse occur due to development of resistance to the therapy. One important cause of resistance to proteasome inhibition is the so-called bounce-back response, a recovery pathway driven by the TCF11/Nrf1 transcription factor, which activates proteasome gene re-synthesis upon impairment of the proteasome function. Thus, inhibiting this recovery pathway potentiates the cytotoxic effect of proteasome inhibitors and could benefit treatment outcomes. DDI2 protease, the 3D structure of which resembles the HIV protease, serves as the key player in TCF11/Nrf1 activation. Previous work found that some HIV protease inhibitors block DDI2 in cell-based experiments. Nelfinavir, an oral anti-HIV drug, inhibits the proteasome and/or pAKT pathway and has shown promise for treatment of relapsed/refractory multiple myeloma. Here, we describe how nelfinavir inhibits the TCF11/Nrf1-driven recovery pathway by a dual mode of action. Nelfinavir decreases the total protein level of TCF11/Nrf1 and inhibits TCF11/Nrf1 proteolytic processing, likely by interfering with the DDI2 protease, and therefore reduces the TCF11/Nrf1 protein level in the nucleus. We propose an overall mechanism that explains nelfinavir's effectiveness in the treatment of multiple myeloma.While traditionally diet quality index scores (DQIS) as noted later in this abstract have been used to predict health outcomes, dietary total antioxidant capacity (TAC), a useful tool for assessing total antioxidant power in the diet, may also be a novel predictor. This study evaluated the associations between dietary TAC and DQIS and all-cause mortality. Based on the National Health and Nutrition Examination Survey (NHANES) 1988-1994 and 1999-2006, 23,797 US adults were followed-up until 2015. Dietary TAC and DQIS including the Healthy Eating Index-2015 (HEI-2015), Alternative Healthy Eating Index-2010 (AHEI-2010), alternate Mediterranean Diet (aMED), and Dietary Approaches to Stop Hypertension (DASH) were calculated using a 1-day 24 h dietary recall. US adults in the highest quintiles of DQIS had lower rates of all-cause mortality compared to those in the lowest quintiles (HEI-2015 hazard ratio (HR) 0.87, 95% confidence interval (CI) 0.77-0.98; AHEI-2010 HR 0.84, 95% CI 0.74-0.94; aMED HR 0.79, 95% CI 0.69-0.90; DASH HR 0.80, 95% CI 0.70-0.92). Similarly, those in the highest quintile of dietary TAC also had a lower all-cause mortality than those in the lowest quintile (HR 0.88, 95% CI 0.79-0.98). These findings suggest that dietary TAC might be a relatively valid predictor of all-cause mortality in the US population compared to the DQIS.Type VI secretion systems (T6SSs) are contractile bacterial multiprotein nanomachines that enable the injection of toxic effectors into prey cells. The Pseudomonas fluorescens MFE01 strain has T6SS antibacterial activity and can immobilise competitive bacteria through the T6SS. Hcp1 (hemolysin co-regulated protein 1), a constituent of the T6SS inner tube, is involved in such prey cell inhibition of motility. Paradoxically, disruption of the hcp1 or T6SS contractile tail tssC genes results in the loss of the mucoid and motile phenotypes in MFE01. Here, we focused on the relationship between T6SS and flagella-associated motility. Electron microscopy revealed the absence of flagellar filaments for MFE01Δhcp1 and MFE01ΔtssC mutants. Transcriptomic analysis showed a reduction in the transcription of class IV flagellar genes in these T6SS mutants. However, transcription of fliA, the gene encoding the class IV flagellar sigma factor, was unaffected. We investigated the magnetic field (H) dependence of magnetization (M) at the Curie temperature TC, and at T = 4.2 K, which concerns the ground state of the ferromagnetic state. The M-H result at TC was analyzed by means of the H versus M5 dependence. At 4.2 K, it was investigated by means of an Arrott plot (H/M vs. M2) according to Takahashi's theory. As for Ni2MnIn and Ni2MnSn, the spin fluctuation parameters in k-space (momentum space, TA) and that in energy space (frequency space, T0) obtained at TC and 4.2 K were almost the same. The average values obtained at TC and 4.2 K were TA = 342 K, T0 = 276 K for Ni2MnIn and TA = 447 K, T0 = 279 K for Ni2MnSn, respectively. The forced magnetostriction at TC was also investigated. The forced linear magnetostriction (ΔL/L) and the forced volume magnetostriction (ΔV/V) were proportional to M4, which followed Takahashi's theory. We compared the forced volume magnetostriction ΔV/V and mechanical parameter, bulk modulus K. ΔV/V is inversely proportional to K. We also discuss the spin polarization of Ni2MnIn and other magnetic Heusler alloys. The pC/pS of Ni2MnIn was 0.860. This is comparable with that of Co2MnGa, which is a famous half-metallic alloy.The use of polymeric nanocomposites has arisen as a promising solution to take advantage of the properties of nanoparticles (NPs) in diverse applications (e.g., water treatment, catalysis), while overcoming the drawbacks of free-standing nanoparticles (e.g., aggregation or accidental release). In most of the cases, the amount and size of the NPs will affect the stability of the composite as well as their performance. Therefore, a detailed characterization of the NPs present on the nanocomposites, including their quantification, is of vital importance for the optimization of these systems. However, the determination of the NPs load is often carried out by destructive techniques such as TGA or ICP-OES, the development of non-invasive approaches to that aim being necessary. In this work, the amount of silver NPs synthesized directly on the surface of melamine (ME) foams is studied using two non-invasive approaches colorimetry and X-ray radiography. The obtained results show that the amount of silver NPs can be successfully determined from the luminosity and global color changes of the surface of the foams, as well as from the X-ray attenuance.
