It is aimed to investigate how intake of high-fat meals composed of different dairy products with a similar fat content affects postprandial peripheral blood mononuclear cell (PBMC) expression of inflammation-related genes, as well as circulating inflammatory markers and metabolites. Healthy subjects (n = 47) consume four different high-fat meals composed of either butter, cheese, whipped cream, or sour cream in a randomized controlled cross-over study. Fasting and postprandial PBMC gene expression, plasma metabolites, and circulating inflammatory markers are measured. Using a linear mixed model, it is found that expression of genes related to lymphocyte activation, cytokine signaling, chemokine signaling, and cell adhesion is differentially altered between the four meals. In general, intake of the fermented products cheese and sour cream reduces, while intake of the non-fermented products butter and whipped cream increases, expression of these genes. Plasma amino acid concentrations increase after intake of cheese compared to the other meals, and the amino acid changes correlate with several of the differentially altered genes. Intake of fermented dairy products, especially cheese, induces a less inflammatory postprandial PBMC gene expression response than non-fermented dairy products. These findings may partly explain inconsistent findings in studies on health effects of dairy products. The state-of-the-art Mie extinction extended multiplicative signal correction (ME-EMSC) algorithm is a powerful tool for the recovery of pure absorbance spectra from highly scatter-distorted spectra. However, the algorithm is computationally expensive and the correction of large infrared imaging datasets requires weeks of computations. In this paper, we present a deep convolutional descattering autoencoder (DSAE) which was trained on a set of ME-EMSC corrected infrared spectra and which can massively reduce the computation time for scatter correction. Since the raw spectra showed large variability in chemical features, different reference spectra matching the chemical signals of the spectra were used to initialize the ME-EMSC algorithm, which is beneficial for the quality of the correction and the speed of the algorithm. One DSAE was trained on the spectra, which were corrected with different reference spectra and validated on independent test data. The DSAE outperformed the ME-EMSC correction in terms of speed, robustness, and noise levels. With intriguing photophysical properties in the red to near-infrared (NIR) regions, facile modulation of their electronic nature by fine-tuning chemical structures, and coordination ability with diverse metal ions, these novel porphyrinoids have favorable prospects in the fields of optical materials, bioimaging and therapy, and catalysis. In this Minireview, we summarize the brief history of porpholactone chemistry, and focus on the studies carried out in our group, particularly on the regioisomeric effect, NIR lanthanide luminescence, and metal catalysis. We outline the perspectives of these compounds in the construction of porpholactone-related biomedical applications and optical and energy materials, in order to inspire more interest and further advance bioinspired inorganic chemistry and lanthanide chemical biology. To predict end-stage renal disease (ESRD) in patients with type 2 diabetes by using machine-learning models with multiple baseline demographic and clinical characteristics. In total, 11 789 patients with type 2 diabetes and nephropathy from three clinical trials, RENAAL (n = 1513), IDNT (n = 1715) and ALTITUDE (n = 8561), were used in this study. Eighteen baseline demographic and clinical characteristics were used as predictors to train machine-learning models to predict ESRD (doubling of serum creatinine and/or ESRD). We used the area under the receiver operator curve (AUC) to assess the prediction performance of models and compared this with traditional Cox proportional hazard regression and kidney failure risk equation models. The feed forward neural network model predicted ESRD with an AUC of 0.82 (0.76-0.87), 0.81 (0.75-0.86) and 0.84 (0.79-0.90) in the RENAAL, IDNT and ALTITUDE trials, respectively. The feed forward neural network model selected urinary albumin to creatinine ratio, serum albumin, uric acid and serum creatinine as important predictors and obtained a state-of-the-art performance for predicting long-term ESRD. Despite large inter-patient variability, non-linear machine-learning models can be used to predict long-term ESRD in patients with type 2 diabetes and nephropathy using baseline demographic and clinical characteristics. The proposed method has the potential to create accurate and multiple outcome prediction automated models to identify high-risk patients who could benefit from therapy in clinical practice. Despite large inter-patient variability, non-linear machine-learning models can be used to predict long-term ESRD in patients with type 2 diabetes and nephropathy using baseline demographic and clinical characteristics. The proposed method has the potential to create accurate and multiple outcome prediction automated models to identify high-risk patients who could benefit from therapy in clinical practice.
