In parallel, opioid abuse has increased and is of major concern. The accessibility of pharmacologically powerful medicines and the Addictovigilance signals in France about the risk of opiates addiction call for an overview of analgesic use. The objective of this study was to investigate the use of analgesics reimbursed in France over a 10-year period through its prevalence. Methods A cross-sectional study repeated yearly was conducted by using data from the French reimbursement database from 2006 to 2015. Analgesics were classified according to their pharmacological potency prevalence of use for each category and socio-demographic characteristics of patients treated were analyzed. Results The annual prevalence of analgesic use was high and increased during the study period (59.8%, 253,976 users in 2015). In 2015, prevalence was always higher in women and increased with age, except for those over 84 years old. Peripheral analgesics were the most used (55.3%, 234,739 users). The prevalence of weak analgesic use decreased (21.3%, 90,257 users), mainly due to the definitive withdrawal of dextropropoxyphene in France in 2011, which was not offset by an increase in the consumption of other weak analgesics. For strong analgesics (1.2%, 5,129 users), morphine was the most widely used with a dramatic increase in oxycodone use, especially in the elderly. Discussion The prevalence of analgesic use is high approximately 31 million adults had at least one analgesic reimbursed in 2015. The most widely used analgesics were peripheral analgesics, far ahead of opioid analgesics.A peak force quantitative nanomechanical mode (PF-QNM) can be used to simultaneously measure the topographical and nanomechanical property maps of samples. Currently, few studies used this mode to investigate the phase characteristics of bitumen in mastics and mixtures. The present study adopted hot-bitumen-pouring methods to prepare base bitumen surface samples and further used frozen-storage and low-temperature-cutting methods to prepare the internal samples of base bitumen in mastics and mixtures. Then, the atomic force microscopy (AFM) PF-QNM mode was used to collect data on the topographical and nanomechanical properties of bitumen and bitumen in mastics and mixtures. The results indicated that a typical bee structure only appeared on the bitumen surface. In contrast to the bitumen surface, the bitumen interior showed two phases, A-phase and B-phase, which were close in some properties to the periphase and paraphase of the bitumen surface, respectively. Furthermore, the addition of mineral aggregates affected the phase characteristics of the free bitumen interior. With an increase in the mineral aggregate surface area, the proportion of A-phase decreased and that of B-phase increased in the free bitumen interior. This article is protected by copyright. All rights reserved.Background and purpose Ethaninidothioic acid (R5421) has been used as a scramblase inhibitor to determine the role of phospholipid scrambling across a range of systems including platelet procoagulant activity. The selectivity of R5421 has not been thoroughly studied. Here, we characterised the effects of R5421 on platelet function and its suitability for use as a scramblase inhibitor. Experimental approach Human platelet activation was measured following pretreatment with R5421 and stimulation with a range of agonists. Phosphatidylserine exposure was measured using annexin V binding. Integrin αIIb β3 activation and α-granule release were measured by flow cytometry. Cytosolic Ca2+ signals were measured using Cal520 fluorescence. An in silico ligand-based screen identified 16 compounds which were tested in these assays. Thiodicarb additionally disrupted Ca2+ homeostasis in unstimulated platelets. Conclusion and implications R5421 is not a selective inhibitor of platelet scramblase activity. We have identified the pesticide thiodicarb, which had similar effects on platelet function to R5421 as well as additional disruption of Ca2+ signalling which may underlie some of thiodicarb's toxicity.Fibrous papule (angiofibroma) is a common benign tumor often presenting as an erythematous papule on the face of an adult, most commonly on the nose. The clinical differential diagnosis is often fibrous papule, basal cell carcinoma, or intradermal nevus. A number of histologic variants of fibrous papule are described, including a variant with junctional melanocytic hyperplasia that can resemble lentigo maligna. These lesions, however, have an unremarkable clinical course and are clinically consistent with a benign fibrous papule. We studied 449 fibrous papules from 425 patients, finding junctional melanocytic hyperplasia noticeable at scanning magnification in 2.9%. Dermatologists and pathologists should be aware that benign fibrous papules can demonstrate junctional melanocytic hyperplasia which can mimic melanoma in-situ.
