Resting frontal EEG alpha asymmetry has been extensively examined as a marker of motivational disposition. Recent research has examined how this trait-level marker of motivation influences an individual's internal error monitoring (indexed by the error-related negativity; ERN), with mixed findings as to whether more negative ERNs are associated with greater left or right alpha power. With the hope that we explored the effect and metabolism of cold plasma (CP) seed treatment on the chilling tolerance in tomato plants. 75 W CP seed treatment showed the best mitigative effect on cold-induced injury of tomato seedlings, as evidenced by the higher maximum photochemical efficiency of PSII (Fv/Fm), lower ion leakage and chilling injury index. Moreover, the results showed that CP-induced chilling tolerance was related to the hydrogen peroxide (H2O2) mediated by respiratory burst oxidase homologue 1 (RBOH1), which was proved by the decrease low temperature tolerance observed in RBOH1 silence or chemical scavenging of H2O2 seedlings. Furthermore, RBOH1-mediated H2O2 acted as the downstream signaling of CP treatment to enhance the levels of abscisic acid (ABA) by increasing the transcript of 9-cis-epoxycarotenoid dioxygenase 1 (NCED1). Mutation of NCED1 completely abolished CP-induced cold resistance. Genetic evidence showed that H2O2 and ABA were positive regulators of cold stress tolerance. Thus, CP-induced H2O2 and ABA cascade signal up-regulated the regulatory genes (ICE1 and CBF1) of cold acclimation, which increased the osmotic adjustment substances (proline and soluble sugar) accumulation and antioxidant enzymes (SOD, APX and CAT) activities. Our results indicate that H2O2 and ABA signals are involved in conferring cold stress tolerance induced by CP seed treatment in tomato plants.Nitric oxide (NO) is a ubiquitous cell signaling molecule which mediates widespread and diverse processes in the cell. These NO dependent effects often involve activation (e.g. NO binding to the heme group of soluble guanylyl cyclase for cGMP production) or inactivation (e.g. S-nitrosation) of protein targets. We studied the effect of NO and heme-NO on the transmembrane signaling enzyme NADPH oxidase 5 (NOX5), a heme protein which produces superoxide in response to increases in intracellular calcium. We found that treatment with NO donors increases NOX5 activity through heme-dependent effects, and that this effect could be recapitulated by the addition of heme-NO. This work adds to our understanding of NOX5 regulation in the cell but also provides a framework for understanding how NO could cause widespread changes in hemeprotein activity based on different affinities for heme v. heme-NO, and helps explain the opposing roles NO plays in activation and inactivation of hemeprotein targets. Chronic rhinosinusitis is an intractable symptom that influences daily lives of patients. miR-1287-5p was discovered to play a suppressive role in cervical cancer and HBV-related infection. This study investigated the potential role of miR-1287-5p in the in-vitro model of chronic rhinosinusitis. GSE169376 dataset was analyzed and differential miRNAs in nasal mucosa tissues in the chronic rhinosinusitis group were screened out. LPS was used to treat HNECs for 12h, 24h and 48h. Cells underwent LPS treatment after SNAI1 downregulation, miR-1287-5p upregulation or pretreatment of the HMGB1 inhibitor, Glycyrrhizin. RT-PCR was used to measure the RNA expression of miR-1287-5p, SNAI1 and HMGB1. ELISA was used for the detection of IL-6, IL-8, TNF-α changes. Targetscan and starBase were used to predict the targets (SNAI1 and HMGB1) of miR-1287-5p. Dual-luciferase reporter assays were applied to validate this. Western blot was used to analyze the protein changes of Snai1, Vimentin, E-cadherin and HMGB1. miR-1287-5p was downregulated in the chronic rhinosinusitis group and decreased after LPS treatment in HNECs. The upregulation of miR-1287-5p inhibited IL-6, IL-8, TNF-α and EMT. miR-1287-5p targeted and inhibited SNAI1 and HMGB1. SNAI1 downregulation led to inhibition in EMT while loss of HMGB1 contributed to the decrease in pro-inflammatory cytokines. Supercritical (sc) sterilization, i.e. the use of scCO2 as a sterilizing agent, emerges as a promising sterilization method due to the mild operational conditions and excellent penetration capability. In this work, a scCO2 protocol was implemented for the one-pot preparation and sterilization of poly(ε-caprolactone) (PCL)/poly(lactic-co-glycolic acid) (PLGA) scaffolds.
