V.ETHNOPHARMACOLOGICAL RELEVANCE Anoectochilus Roxburghii (AR) has been used as a health food and a herb for the curative effect of longevity in China for hundreds years. AIM OF THE STUDY Pharmacological studies demonstrated that the plant ethanol extract possessed anti-oxidative and anti-aging effects on natural aging mice. However, it has not been reported whether the AR flavonoids extract (ARF) has a protective effect on oxidative stress in LO2 cells induced by H2O2 and D-gal induced aging mouse model. MATERIALS AND METHODS In this study, H2O2-induced oxidative stress LO2 cell model was performed to investigate the protective effect of ARF against H2O2 - induced oxidative damage in the LO2 cells by the method of MTT, the content of MDA and SOD, GSH-PX activities were determined. In D-gal induced aging mouse model, we observed behavioral assessment using the Morris water maze (MWM) and pathological morphology of the skin and liver, detected the biomarkers (SOD, GSH-PX, MDA, MAO, AChE) in serum, liver, brain and antioxidant-related gene expression in liver. RESULTS The data showed that ARF had certain protective effect on LO2 cells induced by H2O2 and effectively increased activities of SOD, GSH-PX, and decreased MDA content in cell supernatant.
