Furthermore, several interactions among small-molecule compounds and three G protein-coupled receptor targets (glucagon-like peptide-1 receptor, glucagon receptor, and vasoactive intestinal peptide receptor) predicted using DeepCPI were experimentally validated. The endoplasmic reticulum (ER) is composed of distinct structural domains that perform diverse essential functions, including the synthesis of membrane lipids and proteins of the cell endomembrane system. The polarized growth of fungal hyphal cells depends on a polarized secretory system, which delivers vesicles to the hyphal apex for localized cell expansion, and that involves a polarized distribution of the secretory compartments, including the ER. Here we show that, additionally, the ER of the ascomycete Podospora anserina possesses a peripheral ER domain consisting of highly dynamic pleomorphic ER sub-compartments, which are specifically associated with the polarized growing apical hyphal cells. Apart from spectral analysis, examination of antioxidant ability and protective efficiency showed the differences among newly prepared compounds and commercial antioxidants - gallic acid (GA), methyl gallate (MG), and octyl gallate (OG) applying radical scavenging 1,1-diphenyl-2-picryl-hydrazyl (DPPH), reducing power and iron-chelating assays. The genotoxicity and DNA-protective potential of tested compounds on human peripheral blood mononuclear cells (PBMCs) were evaluated using the single-cell gel electrophoresis (comet assay) and DNA-topology assay. Experimental data revealed that gallotannins G3Rham, G4Man, and G4Glc possess significant radical scavenging/antioxidant activities and manifest very low genotoxic effect on human PBMCs. Moreover, tested compounds considerably reduce the level of DNA damage induced by hydrogen peroxide or Fe2+-ions. The results imply that new synthetic gallotannins can be considered as nontoxic agents for subsequent design of new antioxidants with potential biomedical applications. BACKGROUND Due to the important role of follistatin (FLST), myostatin (MSTN) and growth differentiation factor 11 (GDF11) in muscle mass regulation; alterations in the FLST to MSTN ratio (FM) may result in muscle mass changes in response to different concurrent training (CT) order. This study investigated the influence of 8 weeks of CT order on body composition and serum concentrations of FLST, MSTN, their ratio (FM) and GDF11 in sarcopenic elderly men. METHODS Thirty sarcopenic elderly men (age = 64.3 ± 3.5 years) were randomly assigned into one of three groups, endurance followed by resistance training (E + R; n = 10), resistance followed by endurance training (R + E; n = 10) or control (C; n = 10). Serum concentrations of muscle regulatory markers, body composition, maximum rate of oxygen consumption (VO2max), and upper and lower body strength were evaluated at baseline and after 8 weeks. The training protocol consisted of three training sessions per week for eight weeks. RESULTS There were significant gro+ R = 9.8 kg and R + E = 4.4 kg) significantly increased (P less then 0.01) in the E + R and R + E compared to no changes in the C group. CONCLUSIONS CT increases the FM ratio and FLST as well as reducing MSTN and GDF11 in sarcopenic elderly men. Additionally, CT improved weight, body composition, muscle mass, function, and aerobic fitness. Notably, these results after CT were achieved irrespective of endurance and resistance exercise order in this population. ETHNOPHARMACOLOGICAL RELEVANCE Erythrophleum ivorense (A Chev.) is a common plant in the tropics. Its use as ordeal poison in folklore medicine is controversial. The incoordination and behavioral changes following consumption are often associated with guilt. This study is aimed at dispelling or upholding this belief by investigating the actions of E. ivorense on the brain and behavior using rat model. MATERIALS AND METHODS Sixty male Wistar rats were equally divided into five groups; control group received distilled water, test groups were administered 10, 20, 30 and 40 mg/kg ethanolic extract of E. ivorense in a daily oral dose for 28 days. Cognition (Morris water maze) depression (forced swim test), motor function (hanging wire and inverted wire mesh grid grip tests) and exploratory assessments were done. Brains were stained with H&E, Cresyl violet and immunohistochemistry was done using GFAP, anticalbindin-D28k, Iba-1 and MBP antibodies. RESULTS At all doses, E. ivorense significantly (P ≤ 0.05) increased escape latency in the Morris water maze compared to control. Forced swim test showed a dose-related increase in duration of immobility, significant reduction in hanging latency in hanging wire and wire mesh grid grip test was also observed. Depletion of Purkinje cells of the cerebellum and hippocampal neurons was observed with H&E and cresyl violet. Immuno-staining revealed astrocytic activation in the cerebellum, loss of dendritic spines, cortical microglial activation and demyelination in the cerebellum and dentate gyrus of the hippocampus. CONCLUSION The ethanolic extract of E. ivorense stem bark caused a dose-dependent deficit in learning, memory and motor coordination with evidences of depression in rats. It is concluded that the plant is neurotoxic and induce several neurobehavioral changes.
