However, the molecular feature of SI-NETs remains largely unclear with key genes and pathways yet to be identified. The gene expression profile GSE65286 was retrieved for analysis. Artificial neural networks (ANNs) were constructed for the hub genes network models. A total of 613 differentially expressed genes (DEGs) were identified between normal (N) and primary tumor (T) groups, whereas 61 DEGs were identified between T and liver metastases (LM) groups. The top Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways for the DEGs of N versus T were fat digestion and absorption pathway. For T versus LM the top KEGG pathways were complement and coagulation. In gene set enrichment analysis (GSEA), five gene sets, including Notch signaling, inflammatory response, coagulation, KRAS signaling, and allograft rejection were significantly enriched in the T group. The hub genes in the DEGs of T versus LM included albumin, fibrinogen gamma chain, alpha 2-HS glycoprotein, transferrin and GC, vitamin D binding protein. A distinct correlational alteration of hub genes was observed between T and LM groups. Here, we developed a porous titanium cage by electron beam melting that was filled with simvastatin/poloxamer 407 hydrogel and evaluated bone ingrowth and spinal fusion. Six adult male rhesus macaques underwent interbody fusion of the L3/4 (cage without simvastatin or hydrogel), L4/5 (cage with 0.5 mg simvastatin in hydrogel) and L5/6 (cage with 0 mg simvastatin in hydrogel) segments. 18F-NaF PET/CT imaging confirmed that simvastatin/poloxamer 407 hydrogel increased bone blood flow, osteoblastic activity and bone ingrowth; histological analysis demonstrated that simvastatin/poloxamer 407 hydrogel promoted bone ingrowth in and osseointegration around the L4/5 cages; and biomechanical analysis verified that simvastatin/poloxamer 407 hydrogel enhanced spinal fusion. We conclude that 3D-printed porous cages containing simvastatin hydrogel promote bone ingrowth and spinal fusion, which could be a convenient and promising method for clinical translation. Using PET/CT to monitor the osteoblast activity may provide a new method for assessing the intervertebral fusion effect in preclinical study or clinical postoperative evaluation.A novel molecularly imprinted polymer nanoparticle-based assay (MINA) performed in magnetic microplates was developed as an improved high-quality alternative to existing antibody-based immunoassays. MINA is a generic technology that can be adapted for biomarker detection in biological samples. Herein, we demonstrate the applicability of the MINA assay for the detection of leukotrienes and insulin in biological samples. MINA, used in a competition format, has allowed the detection of LTE4 in urine in a concentration range from 0.45 to 364 pM, with a LOD of 0.73 pM. MINA, used in a competition format, has allowed the detection of insulin in plasma in a concentration range from 25 to 2500 pM, with a LOD of 27 pM. This assay has shown comparable performance for LTE4 and insulin detection to existing chromatographic techniques (LC-MS/MS) and immunoassays in clinically relevant concentrations. The main advantages of this assay are the efficient and low cost fabrication, preparation of synthetic binders without the use of animals, and fewer steps used in the assay protocol as compared to traditional immunoassays.Naked-eye sensing of phytic acid, one of the most abundant antinutrients, was achieved in 100% water medium using a charge transfer complex, composed of pyranine and methyl viologen. Since both the ingredients are commercially available, the design of such sensory systems needs zero synthetic effort, which essentially makes it economically viable. Only the physical mixture of both of these compounds showed a color-changing response from brown to yellow in the presence of phytic acid with a turn-on fluorescence response (LOD 0.56 nM). The electrostatic interaction leads to charge pairing between phytic acid and methyl viologen, which releases free pyranine in solution. Considering its high sensitivity, low-cost test strips were developed for the on-site detection of phytic acid, even in remote locations. Additionally, estimation of phytic acid was achieved in grain samples with a sufficiently high accuracy, as evident from a sufficiently low relative standard deviation ( less then 5%).
