INTRODUCTION Infections associated with medical devices are often related to colonization by Candida spp. biofilm; in this way, numerous strategies have been developed and studied, mainly in order to prevent this type of fungal growth. AIM Considering the above, the main objective of the present study is to make a rational choice of the best antifungal therapy for the in vitro treatment of the biofilm on venous catheters, proposing an innovative formulation of a film-forming system to coat the surface in order to prevent the formation of biofilms. METHODOLOGY Anidulafungin, fluconazole, voriconazole, ketoconazole, amphotericin B, and the association of anidulafungin and amphotericin B were tested against biofilms of C. albicans, C. tropicalis, and C. parapsilosis strains in microtiter plates and in a polyurethane catheter. Besides, anidulafungin, amphotericin B, and the combination of both were incorporated in a film-forming system and were evaluated against biofilm. RESULTS The superior activity of anidulafungin was demonstrated in relation to the other antifungal agents. Although amphotericin B showed good activity, high concentrations were required. The combination showed a synergistic action, in solution and in the formulation, showing excellent results, with activity above 90%. CONCLUSION Due to the superiority of anidulafungin and the synergistic activity of the combination, these alternatives were the most promising options for use in a formulation proposal as a new strategy to combat the Candida spp. biofilm. These formulations demonstrated high in vitro performance in the prevention of biofilms, indicating that they are candidates with great potential for in vivo tests.PURPOSE When considering malaria disease severity, estimation of parasitemia in erythrocytes is important, but sometimes misleading, since the infected erythrocytes may be sequestered in peripheral capillaries. In African children and Asian adults with falciparum malaria, parasitemia as assessed by quantitative PCR (qPCR) in plasma seems to be a valuable indicator of disease severity, but data on African adults as well as the impact of co-infection with HIV is lacking. METHODS In 131 patients with falciparum malaria in a public tertiary teaching hospital in Mozambique, plasma malaria parasitemia as assessed by qPCR, compared to qualitative malaria PCR in blood cell fraction, was related to malaria disease severity and HIV co-infection. https://www.selleckchem.com/products/smip34.html RESULTS Of the 131 patients with falciparum malaria, based on positive qualitative PCR in the blood cell fraction, 93 patients (72%) had positive malaria qPCR in plasma. Patients with severe malaria as defined by the WHO criteria had higher malaria quantitative plasma parasitemia (median 143 genomes/µL) compared to those with uncomplicated malaria (median 55 genomes/µL, p = 0.037) in univariate analysis, but this difference was attenuated after adjusting for age, sex and HIV co-infection (p = 0.055). A quarter of the patients with severe malaria had negative qPCR in plasma. CONCLUSIONS This study of adult African in-patients with falciparum malaria with and without HIV co-infection, neither confirms nor rejects previous studies of malaria qPCR in plasma as an indicator of disease severity in patients with falciparum malaria. However, the specific influence that the structure of one's task representation has on executive functioning has only recently been investigated. Prior research suggests that adjustments of cognitive control are affected by subtle manipulations of aspects of the stimulus-response pairs within and across task sets. This work has focused on examples of cognitive control such as response preparation, dual-task performance, and the congruency sequence effect. The current study investigates the effect of task representation on another example of control, post-error slowing. To determine if factors that influence how people represent a task affect how behavior is adjusted after an error, an adaptive attention-shifting task was developed with multiple task delimiting features. Participants were randomly assigned to a separate task set (two task sets) or an integrated task set (one task set) group. For the separate set group, the task sets switched after each trial. Results showed that only the integrated set group exhibited post-error slowing. This suggests that task representation influences the boundaries of cognitive control adjustments and has implications for our understanding of how control is organized when adjusting to errors in performance.Perception of sounds occurs in the context of surrounding sounds. When spectral properties differ between earlier (context) and later (target) sounds, categorization of later sounds becomes biased through spectral contrast effects (SCEs). Past research has shown SCEs to bias categorization of speech and music alike. Recent studies have extended SCEs to naturalistic listening conditions when the inherent spectral composition of (unfiltered) sentences biased speech categorization. Here, we tested whether natural (unfiltered) music would similarly bias categorization of French horn and tenor saxophone targets. Preceding contexts were either solo performances of the French horn or tenor saxophone (unfiltered; 1 second duration in Experiment 1, or 3 seconds duration in Experiment 2) or a string quintet processed to emphasize frequencies in the horn or saxophone (filtered; 1 second duration). Both approaches produced SCEs, producing more "saxophone" responses following horn / horn-like contexts and vice versa. One-second filtered contexts produced SCEs as in previous studies, but 1-second unfiltered contexts did not. Three-second unfiltered contexts biased perception, but to a lesser degree than filtered contexts did. These results extend SCEs in musical instrument categorization to everyday listening conditions.
