t.BACKGROUND Opioid agonist activation at the mu opioid receptor (MOR) can lead to a wide variety of physiological responses. Many opioid agonists share the ability to selectively and preferentially activate specific signaling pathways, a term called biased agonism. Biased opioid ligands can theoretically induce specific physiological responses and might enable the generation of drugs with improved side effect profiles. METHODS Dynorphins, enkephalins, and endomorphins are endogenous opioid agonist peptides that may possess distinct bias profiles; biased agonism of endogenous peptides could explain the selective roles of these ligands in vivo. Our purpose in the present study was to investigate biased signaling and potential underlying molecular mechanisms of bias using 35S-GTPγS and cAMP assays, specifically focusing on the role of adenylyl cyclases (ACs) and regulators of G-protein signaling proteins (RGSs) in CHO, N2a, and SH-SY5Y cell lines, all expressing the human MOR. RESULTS We found that endomorphin-1/2 preferentially activated cAMP signaling, while dynorphin-B preferentially activated 35S-GTPγS signaling in most cell lines. Experiments carried out in the presence of an isoform selective RGS-4 inhibitor, and siRNA knockdown of AC6 in N2a cells did not significantly affect the bias properties of endomorphins, suggesting that these proteins may not play a role in endomorphin bias. CONCLUSION We found that endomorphin-1/2 and dynorphin-B displayed contrasting bias profiles at the MOR, and ruled out potential AC6 and RGS4 mechanisms in this bias. METHODS An electronic search of PubMed was combined with an extended search of references within systematic reviews and expert suggestions. Studies were included if they reported on an infant or child under 18 years with exposure to medications; assessed safety or effectiveness; specified a comparison or control group, and were published in English in 2016. Data extraction was conducted by one team member using a standardized form and reviewed by a second team member. Study quality was assessed using the GRACE checklist for rating the quality of observational studies. RESULTS After removing study medication safety and effectiveness in children. Our systematic review appears generalizable to pediatrics broadly, and documents that the high level of activity in RWE in general has had less of an impact on pediatrics.In recent years, with the frequent occurrences of heavy metal (HM) pollution in agriculture, the problem of HM pollution in farmland soil, especially in the areas irrigation by the Yellow River, has been attracted increasing attention because of the complex sources of pollution. Qualitative identification of pollution sources and quantification of their contributions to HMs in soil are the key links in the prevention and control of HM pollution. The contents of 11 heavy metals (As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Sn, V, and Zn) in the rhizosphere soil of the Ningxia irrigation area were determined by inductively coupled plasma mass spectrometry (ICP-MS). Multiple methods were used for source identification, including positive matrix factorization (PMF) analysis combined with multiple other analyses (single factor index method (Pi), coefficient of variation(CV), correlation analysis(CA), enrichment factor(EF), and principal component analysis(PCA)). The results showed that (1) the over-standard rates of As, Cd, Cr, Mn, Pb, Sn, and Zn in the study area were 100%, of which Cd was seriously polluted, while As, Zn, and Sn were moderately polluted. (2) The HM contributions from irrigation and silt soil formed by the Yellow River sediment were the highest (42.45%), followed by the smelting industry and traffic pollution (16.06%). (3) The contribution of agricultural pollution to HMs in the region was 15.54%, in which As was mainly from pesticides and Cd was mainly from fertilizers.The effects of coagulant dosage, alkalinity, turbidity, ionic strength, and dissolved organic matter (DOM) on the removal of sulfadimethoxazole (SMZ) and oxytetracycline (OTC) by coagulation were studied and the reaction mechanisms of the coagulation process were revealed in this research. From our results, alkalinity, turbidity, ionic strength, and DOM had different effects on the removal of antibiotics. The SMZ and OTC removals were improved with increase in poly-aluminum chloride (PACl) dosage, whereas the turbidity had less influence on the removal of SMZ and OTC because the adsorption of SMZ and OTC to kaolin was low, confirmed by a control when no PACl added.
