The formation of subgel (so-called hydrated crystal) phase of membranes of ether-linked phospholipids, dialkylphosphatidylcholines containing linear saturated alkyl chain (Cn = 14, 16 and 18), was examined under atmospheric and high pressure. The results of differential scanning calorimetry in 50 wt% aqueous ethylene glycol solution and water showed that these PC membranes undergo the subtransition from the subgel phase to the gel phase at a low temperature with or without the thermal pretreatment of lipid samples called annealing. The subtransition in water was clearly observed by light-transmittance measurements under high pressure and the transition temperature increased by applying pressure. The temperature-pressure phase diagrams and the thermodynamic quantities of the subtransition were obtained from the phase-transition data and compared with those of membranes of ester-linked phospholipids, diacylphosphatidylcholines. The phase diagrams indicated that all gel phases of the ether-linked PC membranes exist as stable phases while parts of the gel phases of the ester-linked PC membranes are metastable. The subtransition temperatures of the ether-linked PC membranes were lower than those of the ester-linked PC membranes by more than 10 °C and the corresponding thermodynamic quantities were extremely small. Further, it was revealed by high-pressure fluorometry that the difference in subgel phase between ether- and ester-linked PC membranes results from their phase structures the nonbilayer interdigitated structure is maintained after the conversion from the gel phase to the subgel phase in the ether-linked PC membranes whereas the ester-linked PC membranes form the bilayer subgel phase with staggered structure.Previous studies have shown that melatonin (MT) can ameliorate vitrification-inflicted damage in mouse germinal vesicle (GV) oocytes, however, the key mechanistic basis of this improvement still remains poorly understood. This study was conducted to investigate whether MT can improve in vitro developmental potential of vitrified-warmed GV oocytes through its receptors. The fresh oocytes were randomly divided into four groups untreated (control group, F), vitrified by open-pulled straw method (vitrification group, V), vitrification group with 100 nmol/L MT supplementation (vitrification + MT group, VM), and with 100 nmol/L MT plus 100 nmol/L luzindole administration (vitrification + MT + luzindole group, VML) or with 50 nmol/L ramelteon addition (vitrification + ramelteon group; VR). After warming, oocytes were cultured in vitro, and MT receptors (MTRs), MAD2 (mitotic arrest deficient 2), Securin and CyclinB1 protein levels and spindle morphology were evaluated. The ratio of oocytes developed to the metaphase of the present study shows that MT (100 nmol/L) can ameliorate the in vitro maturation of vitrified-warmed mouse GV oocytes, potentially by improving the spindle morphology, modulating MAD2 protein level and promoting the development of MI stage oocytes through MTRs.Prenatal alcohol exposure (PAE) may result in Fetal Alcohol Spectrum Disorders (FASD). The hippocampus has been recognized as a vulnerable target to alcohol-induced developmental damage. However, the effect of prenatal exposure to alcohol on dendritic morphological adaptations throughout the hippocampal fields in the developing brain still remains largely unknown in the context of FASD. We hypothesized that chronic binge alcohol exposure during pregnancy alters dendrite arborization throughout the developing rat hippocampus. Pregnant Sprague-Dawley rats were assigned to either a pair-fed control (PF-Cont) or a binge alcohol (Alcohol) treatment group. Alcohol dams were acclimatized via a once-daily orogastric gavage of 4.5 g/kg alcohol from gestational day (GD) 5-10 and progressed to 6 g/kg alcohol from GD 11-21. Pair-fed dams similarly received isocaloric maltose dextrin. After parturition, all dams received an ad libitum diet and nursed their offspring until postnatal day (PND) 10 when the pup brains were collected for morphological analysis. PAE increased dendritic arborization and complexities of CA1, CA2/3, and DG neurons in the PND 10 rat hippocampus. The number of primary dendrites, total dendritic length, and number of dendritic branches were significantly increased following PAE, and Sholl analysis revealed significantly more intersections of the dendritic processes in PND 10 offspring following PAE compared with those in the PF-Cont group. We conclude that chronic binge PAE significantly alters hippocampal dendritic morphology in the developing hippocampus. We conjecture that this morphological alteration in postnatal rat hippocampal dendrites following chronic binge prenatal alcohol exposure may play a critical role in FASD neurobiological phenotypes.During the present COVID-19 pandemic, transplantation of donor lungs with a history of COVID-19 infection is a critical issue. Donor-derived virus infection and graft dysfunction are possible after transplantation. However, use of such lungs could save the lives of patients requiring emergency transplantation. We successfully transplanted lungs from a brain-dead donor who had recovered from SARS-CoV-2 into a severe respiratory failure patient supported with ECMO who needed an emergency transplant. At the 3-month follow-up, our patient showed no evidence of COVID-19 transmission or graft dysfunction.
