This study aimed to explore novel microRNAs in plasma for predicting chemoresistance in adjuvant chemotherapy for patients with gastric cancer (GC). We used the Toray 3D-Gene microRNA array-based approach to compare preoperative plasma microRNA levels between GC patients with and without recurrences after curative gastrectomy. All patients underwent adjuvant chemotherapy with S-1, an oral fluoropyrimidine. Of 2566 candidates, six candidate microRNAs (miR-1229-3p, 1249-5p, 762, 711, 1268a and 1260b), which were highly expressed in the preoperative plasma of patients with subsequent recurrences, were selected. In a large-scale validation analysis by quantitative RT-PCR, we focused on high plasma levels of miR-1229-3p, which was an independent poor prognostic factor for recurrence free survival (P = 0.009, HR = 3.71). Overexpression of miR-1229-3p in GC cells induced significant chemoresistance to 5-fluorouracil (5-FU), up-regulation of thymidylate synthase (TS) and dihydroprimidine dehydrogenase (DPD) and down-regulation of SLC22A7 both in vitro and in vivo. Intraperitoneal injection of miR-1229-3p in mice induced significant chemoresistance to 5-FU, accompanied by high levels of miR-1229-3p in plasma and tumor tissue. These findings suggest that plasma miR-1229-3p might be a clinically useful biomarker for predicting chemoresistance to S-1 and selecting other or combined intensive chemotherapy regimens in GC patients.One of the characteristics of global climate change is the increase in extreme climate events, e.g., droughts and floods. Forest adaptation strategies to extreme climate events are the key to predict ecosystem responses to global change. Severe floods alter the hydrological regime of an ecosystem which influences biochemical processes that control greenhouse gas fluxes. We conducted a flooding experiment in a mature grey alder (Alnus incana (L.) Moench) forest to understand flux dynamics in the soil-tree-atmosphere continuum related to ecosystem N2O and CH4 turn-over. The gas exchange was determined at adjacent soil-tree-pairs stem fluxes were measured in vertical profiles using manual static chambers and gas chromatography; soil fluxes were measured with automated chambers connected to a gas analyser. The tree stems and soil surface were net sources of N2O and CH4 during the flooding. Contrary to N2O, the increase in CH4 fluxes delayed in response to flooding. Stem N2O fluxes were lower although stem CH4 emissions were significantly higher than from soil after the flooding. Stem fluxes decreased with stem height. Our flooding experiment indicated soil water and nitrogen content as the main controlling factors of stem and soil N2O fluxes. The stems contributed up to 88% of CH4 emissions to the stem-soil continuum during the investigated period but soil N2O fluxes dominated (up to 16 times the stem fluxes) during all periods. Conclusively, stem fluxes of CH4 and N2O are essential elements in forest carbon and nitrogen cycles and must be included in relevant models.There is a growing awareness of the importance of indoor microbiomes for human health. Given their complexity, these microbiomes can only be adequately surveyed using high throughput sequencing techniques. Oxford Nanopore's MinION is the newest third generation sequencing technology on the market. With its many advantages such as portability, user friendliness, simplicity, speed of sequencing and long read length, the technology is now an actual contender to established sequencing platforms. MinION's main disadvantage is a relatively low read accuracy compared to several other platforms, although this is constantly improving. The present study, which appears to be the first of its kind, provides the results of a preliminary analysis of the microbial communities in indoor environments based on 16S rRNA gene amplicon sequencing, using both the Oxford Nanopore Technologies (ONT) MinIOn and the Illumina MiSeq DNA sequencers. At the level of family and above, there was no significant difference between the microbial compositions as revealed by the two platforms. In this paper, the water activity coefficient of this binary system was measured from 303 K to 363 K with water concentration from 18% to 92%. The interaction energies between the ionic liquid molecules ([Formula see text]) and between the ionic liquid and water molecules ([Formula see text]) for the [EMIM][MeSO3]/water binary system were determined from the water activity coefficient data using the Non-Random Two-Liquid (NRTL) model. The magnitude of the interaction energy between the [EMIM][MeSO3] and water molecules ([Formula see text]) was found to be in the range of 45~49 kJ/mol, which was about 20% larger than that between the water molecules ([Formula see text]) in the [EMIM][MeSO3]/water system. The large ([Formula see text]) can explain many observed macroscopic thermodynamic properties such as strong hygroscopicity in the ionic liquid [EMIM][MeSO3].
