Calcitriol, a vitamin D3 metabolite, is approved for various indications because it is the bioactive form of vitamin D in the body. The purpose of this study was to predict the clinical significance of cytochrome P450 (CYP) induction by calcitriol using in vitro human cryopreserved hepatocytes, HepaRG experimental systems, and various pharmacokinetic estimation models. CYP2B6, 3A4, 2C8, and 2C9 mRNA levels increased in a concentration-dependent manner in the presence of calcitriol in human cryopreserved hepatocytes and HepaRG cells. Using the half maximal effective concentration (EC50) and maximum induction effect (Emax) obtained from the in vitro study, a basic kinetic model was applied, suggesting clinical relevance. In addition, a static mechanistic model showed the improbability of a clinically significant effect; however, the calculated area under the plasma concentration-time curve ratio (AUCR) was marginal for CYP3A4 in HepaRG cells. To clarify the effect of CYP3A4 in vivo, physiologically based pharmacokinetic (PBPK) modeling was applied as a dynamic mechanistic model, revealing a low clinically significant effect of CYP3A4 induction by calcitriol. Therefore, we conclude that CYP induction by calcitriol treatment would not be clinically significant under typical clinical conditions. The innovative work behavior (IWB) or creativity of employees is regarded as the key to the sustainable innovation performance of an organization. In the field of human resource management (HRM), the relationship between an organization's high-performance work system (HPWS) and IWB has been studied extensively. However, the current understanding of organizational external antecedents is limited. Our paper focuses on an extra-organizational government support factor, government support for talent policy (GSTP). Similar to HRM policies within an organization, GSTP also has an ability-motivation-opportunity framework that may influence the IWB of employees. We integrate the resource dependence theory, institutional theory, and theory of planned behavior (TPB) to propose a theoretical model of the mechanism of GSTP influence on IWB. Using a structural equation modeling approach, we empirically verify the hypotheses in a survey dataset of HRM practitioners in 152 technology-based enterprises in China. The results indicate that the external antecedent, GSTP, positively influences the innovative attitude, subjective norm, and perceived behavioral control of HRM practitioners in the organization. Innovative attitude and perceived behavioral control completely mediated the relationship between GSTP and innovative intention. Moreover, there is a distal indirect effect between GSTP and IWB. The paper contributes to filling a gap in the innovation policy literature. In practice, both HPWS and individual employees should be concerned about the possible role of GSTP.According to the basic principle of dense packing of particles, and considering the interaction between particles, a dense packing model of granular materials in concrete was proposed. During the establishment of this model, binary particle packing tests of crushed stone and sand were carried out. The fitting analysis of the test results determines the relationship between the particle size ratio and the remaining volume fraction of the particle packing, and then the actual void fraction of the particle packing was obtained, based on which the water-binder ratio was combined to determine the amount of various materials in the concrete. The proposed concrete mix design method was used to prepare concrete, and its compressive strength and elastic modulus were tested experimentally. The test results show that the aggregate volume fraction of the prepared concrete increased, and the workability of the concrete mixture with the appropriate amount of water reducing agent meets the design requirements. When the water-binder ratio was 0.42, 0.47, or 0.52, the compressive strength of the concrete increased compared with the control concrete, and the degree of improvement in compressive strength increased with the decrease in water-binder ratio; when the water-binder ratio was 0.42, 0.47, or 0.52, the static elastic modulus of the concrete increased compared with the control concrete, and the degree of improvement in elastic modulus also increased with the decrease in water-binder ratio. The elastic modulus and compressive strength of the prepared concrete have a positive correlation. Findings show that the concrete mix design method proposed by this research is feasible and advanced in a sense.Bio-nanotechnology employing bio-sourced nanomaterial is an emerging avenue serving the field of fish medicine. Marine-sourced chitosan nanoparticles (CSNPs) is a well-known antimicrobial and immunomodulatory reagent with low or no harm side effects on fish or their human consumers. In this study, in vitro skin mucus and serum antibacterial activity assays along with intestinal histology, histochemical, and gene expression analyses were performed to evaluate the impact of dietary CSNPs (5 g kg-1 dry feed) on rainbow trout resistance against 'enteric redmouth' disease. Two treatment conditions were included; short-term prophylactic-regimen for 21 days before the bacterial challenge, and long-term therapeutic-regimen for 21 days before the challenge and extended for 28 days after the challenge. https://www.selleckchem.com/products/iwp-2.html Our results revealed higher antibacterial defense ability and positive intestinal histochemical and molecular traits of rainbow trout after dietary CSNPs. The prophylactic-regimen improved trout health while the therapeutic regimen improved their disease resistance and lowered their morbidity. Therefore, it is anticipated that CSNPs is an effective antibacterial and immunomodulatory fish feed supplement against the infectious threats. However, the CSNPs seem to be more effective in the therapeutic application rather than being used for short-term prophylactic applications. The objectives in this study are to improve the performance of PVDF membrane by incorporating TiO2 and silane at various dosages and optimize fabricating conditions by using response surface methodology (RSM) for membrane distillation (MD) application. The PVDF membrane was synthesized by phase inversion method using various TiO2, silane and polymer concentrations. Membranes were characterized by performing contact angle measurements, SEM and FTIR observations. Ammonia rejection and permeate flux were measured by operating a direct contact distillation module treating ammonium chloride solution. A PVDF membrane created by adding TiO2 modified by silane improved membrane hydrophobicity. However, the effect of silane on membrane hydrophobicity was less pronounced at higher TiO2 concentrations. Highest ammonium rejection was associated with the highest membrane hydrophobicity. RSM analysis showed that fabricating conditions to achieve highest flux (10.10 L/m2·h) and ammonium rejection (100.0%) could be obtained at 31.
