MicroRNAs (miRNAs) are small non-coding RNAs with important roles in diverse biological processes including immunity. Japanese flounder (Paralichthys olivaceus) is an aquaculture fish species susceptible to the infection of bacterial and viral pathogens including Edwardsiella tarda. In a previous study, pol-miR-novel_547, a novel miRNA of flounder with unknown function, was found to be induced by E. tarda. In the present study, we investigated the regulation and function of pol-miR-novel_547 and its target gene. We found that pol-miR-novel_547 was regulated differently by E. tarda and the viral pathogen megalocytivirus, and pol-miR-novel_547 repressed the expression of PTEN (phosphatase and tensin homolog) of flounder (PoPTEN). PoPTEN is ubiquitously expressed in multiple tissues of flounder and responded to bacterial and viral infections. Interference with PoPTEN expression in flounder cells directly or via pol-miR-novel_547 promoted E. tarda invasion. Consistently, in vivo knockdown of PoPTEN enhanced E. tarda dissemination in flounder tissues, whereas in vivo overexpression of PoPTEN attenuated E. tarda dissemination but facilitated megalocytivirus replication. Further in vitro and in vivo studies showed that PoPTEN affected autophagy activation via the AKT/mTOR pathway and also modulated the process of apoptosis. Together these results reveal for the first time a critical role of fish PTEN and its regulatory miRNA in pathogen infection, autophagy, and apoptosis.Purpose of the study Work addiction risk is a growing public health concern with potential deleterious health-related outcomes. Perception of work (job demands and job control) may play a major role in provoking the risk of work addiction in employees. We aimed to explore the link between work addiction risk and health-related outcomes using the framework of job-demand-control model. Methods Data were collected from 187 out of 1580 (11.8%) French workers who agreed to participate in a cross-sectional study using the WittyFit software online platform. The self-administered questionnaires were the Job Content Questionnaire by Karasek, the Work Addiction Risk Test, the Hospital Anxiety and Depression scale and socio-demographics. Results There were five times more workers with a high risk of work addiction among those with strong job demands than in those with low job demands (29.8% vs. 6.8%, p = 0.002). This is a paradigm shift, where anything and everything can be interconnected via a communication medium. In such systems, security is a prime concern and protecting the resources (e.g., applications and services) from unauthorized access needs appropriately designed security and privacy solutions. Building secure systems for the IoT can only be achieved through a thorough understanding of the particular needs of such systems. The state of the art is lacking a systematic analysis of the security requirements for the IoT. Motivated by this, in this paper, we present a systematic approach to understand the security requirements for the IoT, which will help designing secure IoT systems for the future. In developing these requirements, we provide different scenarios and outline potential threats and attacks within the IoT. Based on the characteristics of the IoT, we group the possible threats and attacks into five areas, namely communications, device/services, users, mobility and integration of resources. We then examine the existing security requirements for IoT presented in the literature and detail our approach for security requirements for the IoT. We argue that by adhering to the proposed requirements, an IoT system can be designed securely by achieving much of the promised benefits of scalability, usability, connectivity, and flexibility in a practical and comprehensive manner.Magnesium alloys are concerned for its mechanical properties and high damping performance. The influence of Mn toward the internal organization morphology of long-period stacking ordered (LPSO) second phase and the consistent damping performance in Mg-4.9Zn-8.9Y-xMn have been studies in this work. It has shown that the addition of Mn tends to diffuse to the LPSO interface and causes the LPSO phase to expand in the arc direction. The circular structure of LPSO can optimize the damping property of the alloy better than the structure with strong orientation, especially at the strain of 10-3 and 250 °C. With more additions of Mn, damping would have a reduction due to the dispersed fine LPSO phases and α-Mn particles. When the Mn content is higher than 1.02%, the grain is refined, and mechanical properties have been significantly improved. Mg-4.9%Zn-8.9%Y-1.33%Mn shows the best mechanical property.(1) Background Regular exercise induces physiological and morphological changes in the organisms, but excessive training loads may induce damage and impair recovery or muscle growth. The purpose of the study was to evaluate the impact of Silymarin (SM) consumption on endurance capacity, muscle/cardiac histological changes, bodyweight, and food intake in rats subjected to 60 min of regular exercise training (RET) five days per week. (2) Methods Male Wistar rats were subjected to an eight-week RET treadmill program and were previously administered SM and vitamin C. Bodyweight and food consumption were measured and registered. The maximal endurance capacity (MEC) test was performed at weeks one and eight. After the last training session, the animals were sacrificed, and samples of quadriceps/gastrocnemius and cardiac tissue were obtained and process for histological analyzes. (3) Results SM consumption improved muscle recovery, inflammation, and damaged tissue, and promoted hypertrophy, vascularization, and muscle fiber shape/appearance.
