The European AIDS Clinical Society (EACS) Guidelines cover key aspects of HIV management with major updates every twoyears. The 2019 Guidelines were extended with a new section focusing on drug-drug interactions and other prescribing issues in people living with HIV (PLWH). The recommendations for treatment-naïve PLWH were updated with four preferred regimens favouring unboosted integrase inhibitors. A two-drug regimen with dolutegravir and lamivudine, and a three-drug regimen including doravirine were also added to the recommended initial regimens. Lower thresholds for hypertension were expanded to all PLWH and for cardiovascular disease prevention, the 10-year predicted risk threshold for consideration of antiretroviral therapy (ART) modification was lowered from 20% to 10%. Frailty and obesity were added as new topics. It was specified to use urine albumin to creatinine ratio to screen for glomerular disease and urine protein to creatinine ratio for tubular diseases, and thresholds were streamlined witn and are translated into Chinese, French, German, Japanese, Portuguese, Russian and Spanish. Advanced histological stage is an important factor in individual risk stratification in patients with primary biliary cholangitis (PBC). Non-invasive biomarkers for advanced histological stage are needed. We assessed the utility of Gas6 and Axl as biomarkers for advanced histological stage in patients with PBC. A total of 113 biopsy-proven PBC patients and 20 healthy controls were included in this study. Serum Axl and Gas6 were measured using enzyme-linked immunosorbent assay. The Gas6/albumin ratio and Axl/albumin ratio were also evaluated as biomarkers of histological stage. Serum Axl (42.6ng/mL vs. 30.6ng/mL, P < 0.001) and Gas6 (21.1ng/mL vs. 18.8ng/mL, P = 0.007) levels in PBC patients were significantly higher than those in healthy controls. The Axl/albumin ratio was 10.4, and the Gas6/albumin ratio was 7.6 in patients with PBC. Gas6 and Axl were significantly correlated with aspartate aminotransferase, bilirubin, albumin, and platelets. Two challenges that limit sustainable aromatics production via Diels-Alder (DA) aromatisation-an unfavourable equilibrium position and undesired regioselectivity when using asymmetric addends-were addressed using a dynamic kinetic trapping strategy. Activated acrylates were used to speed up the forward and reverse DA reactions, allowing for one of the four DA adducts to undergo a selective intramolecular lactonisation reaction in the presence of a weak base. The adduct is removed from the equilibrium pool, pulling the system completely to the product with a fixed, desired regiochemistry. A single 1,2-regioisomeric lactone product was formed in up to 86 % yield and the acrylate activating agent liberated for reuse. The lactone was aromatised to give phthalide in almost quantitative yield in the presence of Ac2 O and a catalytic amount of strong acid, or in 79 % using only catalytic acid.Hydrogen-bonded organic frameworks (HOFs) possess various merits, such as high porosity, tunable structure, facile modification, and ready regeneration. These properties have yet to be explored in the context of new functional HOF materials. The facile and inexpensive electrophoretic deposition (EPD) method applied in this study generated a transparent HOF film at room temperature in just 2 min and is applicable to other HOFs. The resulting film exhibited reversible electrochromism with the advantage of long cycle life (>500 cycles). More strikingly, this all-organic film could be readily regenerated (through rinsing with DMF and redeposition) and showed tunable electrochromic behavior (through low-cost postsynthetic modification) with the ability to undergo successive color changes, which is difficult to achieve with conventional electrochromic materials. An electrochromic device was manufactured to further demonstrate the application potential of the film.The filamentous fungus Aspergillus oryzae has 27 putative iterative type I polyketide synthase (PKS) gene clusters, but the secondary metabolites produced by them are mostly unknown. Here, we focused on eight clusters that were reported to be expressed at relatively high levels in a transcriptome analysis. By comparing metabolites between an octuple-deletion mutant of these eight PKS gene clusters and its parent strain, we found that A. oryzae produced 2,4'-dihydroxy-3'-methoxypropiophenone (1) and its precursor, 4'-hydroxy-3'-methoxypropiophenone (3) in a specific liquid medium. Furthermore, an iterative type I PKS (PpsB) encoded by AO090102000166 and an acetyl-CoA ligase (PpsA) encoded downstream from ppsB were shown to be essential for their biosynthesis. PpsC, encoded upstream from ppsB, was shown to have 3-binding activity (Kd =26.0±6.2 μM) and is suggested to be involved in the conversion of 3 to 1. This study deepens our understanding of cryptic secondary metabolism in A. oryzae.Laminin α chains (α1-α5 chains) are expressed in a tissue- and developmental stage-specific manner and have diverse chain-specific biological functions. Especially, laminin globular (LG) modules (LG1-LG5) located at the C-terminus of the α chains play a critical role in the biological activities of laminins. Each LG module is composed of a 14-stranded β-sheet (A-N) sandwich structure. We previously screened cell attachment activity of the loop regions between the E and F strands in the LG modules using 17 homologous peptides (EF peptides) and found that four active EF peptides bind to integrin α2β1. One of the four peptides, G4EF1 demonstrated improved cell attachment activity when cyclized. Here, we focused on the remaining three integrin α2β1-binding EF peptides (G5EF1, G3EF3, and G5EF5) and analyzed the relationship between their peptide conformation and cell attachment activity. First, we determined their active core sequences and found that G5EF1z (IGLEIVDGKVLFHVNN), G3EF3z (LLVTLEDGHIALST), and G5EF5z (KVLTEQVL) are the core sequences.
