Immune checkpoint inhibitors (ICIs) have become one of the standard treatment options for solid tumours; however, treatment-related adverse events, including pneumonitis, sometimes disrupt the ongoing treatment. To evaluate the impact of ICI addition on the risk of pneumonitis, a meta-analysis was conducted. Phase III randomised controlled trials comparing ICIs and conventional therapy with conventional therapy alone were selected by searching databases, including PubMed, Embase, Web of Scienceand Cochrane Library. The odds ratio (OR) of any grade and grade III-V pneumonitis was calculated. Meta-analysis was performed to compare the incidence of pneumonitis between the treatment arm with additional ICIs and the arm with conventional therapy. A total of 25 randomised controlled trials (RCTs) representing 16,343 patients for grade I-V and 23 RCTs with 15,006 patients for grade III-V pneumonitis were analysed. Adding ICIs was associated with a significant increase in pneumonitis (grade I-V OR, 2.67, 95% confidence interval [CI] 2.12-3.37, grade III-V OR, 1.83, 95% CI 1.26-2.65). In subgroup analyses based on the mechanism of action of ICIs, cancer types (lung and non-lung cancer) and each ICI, no significant difference in heterogeneity was observed. Adding ICIs to the conventional treatment for solid tumours significantly increased both grade I-V and grade III-V pneumonitis regardless of the mechanisms of ICIs and cancer type. These results would be helpful for oncologists to choose the appropriate treatment options using ICIs, particularly for patients with known risk factors of pneumonitis or interstitial lung disease. To systematically review randomized trials of the effectiveness of inpatient Consultation-Liaison (C-L) Psychiatry service models in improving patient outcomes, reducing length of hospital stay and decreasing healthcare costs. We searched databases including Ovid Medline, Ovid Embase, Ovid PsycINFO and EBSCO CINAHL for relevant trials. Two independent reviewers assessed articles and extracted data. The review is registered with PROSPERO, number CRD42019120827. Eight trials were eligible for inclusion. All had methodological limitations and all were published more than ten years ago. None reported clear evidence that the C-L Psychiatry service model evaluated was more effective than usual medical care alone. All the service models tested focused on providing a consultation for patients identified by screening. Clinical heterogeneity precluded meta-analysis. The present study investigated the effects of maternal aging on mitochondrial DNA (mtDNA) copy number in embryos, and the amount of cell-free mtDNA (cf-mtDNA) in spent culture medium (SCM) of embryos. Moreover, we examined the effects of vitrification on mtDNA copy number in embryos of young and aged cows, and on cf-mtDNA abundance in SCM. Oocytes collected from ovaries of young (20-40 months old) and aged cows (> 140 months old) were used to produce early stage embryos (8-12 cell-stage, 48 h after insemination). These embryos were individually cultured for 5 days, and mtDNA copy number in blastocysts and cf-mtDNA content in SCM, were evaluated by real-time PCR. At 48 h post-insemination, mtDNA copy number in embryos was greater for young cows compared with that of aged cows, whereas no significant difference was observed in cf-mtDNA in the SCM. Next, we addressed whether zona pellucida (ZP) may mask the difference in cf-mtDNA content in SCM. Using ZP-free embryos, we found significantly greater cf-mtDNA content in the SCM of blastocysts derived from aged cows. Furthermore, when embryos were vitrified and warmed, mtDNA copy number in blastocysts derived from young cows was lower, whereas cf-mtDNA content in SCM was greater than in those derived from aged cows. In conclusion, maternal aging affects mitochondrial kinetics and copy number in embryos following vitrification.Immunohistochemical analysis is a routine procedure for clinical and research studies in male fertility. However, most of the interpretations remain subjective and time-consuming, with inherent intra- and inter-observer variability. Given the prognostic and research implications of testicular assessment, a more objective and less time-consuming method is required. In the current study, we used in vitro matured pre-pubertal murine testes as a model. The main objective was to develop an affordable automated digital immunohistochemistry image analysis tool for an unbiased and quantitative assessment of testicular tissue sections. Testicular explants were fixed, cut, and stained for specific germ cell markers. The classical manual counting procedure was evaluated. Background and noise were reduced on brightfield images. Photomicrographs were stitched (Background_Elimination_Stitching) to create high-quality images. Two procedures were evaluated (IHC_Tool and Stained_Nuclear_Area); then a procedure (Necrotic_Area_Elimination) allowing withdrawal of the necrotic area observed after culture was assessed.
