The performance of each machine learning classifier based on McDSL, LASSO, and Cox was not significantly different. Logistic regression yielded the optimal performance in predicting 3-year PFS based on the McDSL (AUC, 0.872, 95% confidence interval [CI] 0.828-0.916) and 3-year OS based on the LASSO (AUC, 0.901, 95% CI 0.861-0.940). Conclusions McDSL is more reproducible than LASSO and Cox model in the feature selection process. Lysosomal cysteine protease-cathepsin S (CTSS), has been reported to be involved in invasive growth and distant metastasis of cancer cells. However, the underlying mechanisms remained elusive. The CTSS inhibitor-Z-FL-COCHO (ZFL), could attenuate TGF-β-induced invasive growth as proven by wound healing and transwell assays. Furthermore, inhibition of CTSS could reverse TGF-β-induced epithelial-to-mesenchymal transition (EMT) and restore TGF-β-triggered tight junction proteins turnover, thus decreasing glioblastoma cell mobility. We also observed that TGF-β could change the morphology of glioblastoma cells, redistribute intermediate-filament, vimentin, which was highly relevant to mesenchymal type cells and enhanced mobility. However, inhibition of CTSS could significantly restore this transformation. Our results proved that PI3K/AKT/mTOR pathway was significantly suppressed in the TGF-β+ZFL (CTSS inhibitor) groups, and AKT activator-SC79, could reverse the anti-invasion effect of CTSS, indicating an important role of PI3K/AKT/mTOR pathway in this process. Conclusion Z-FL-COCHO (ZFL), a CTSS inhibitor, could reverse TGF-β-induced EMT and change of tight junction proteins via PI3K/AKT/mTOR pathway.Background This study aimed to clarify the relationship between F. nucleatum levels and the prognosis of CRC, which is still controversial. Methods Relevant articles were searched on PubMed, Web of Science, PMC and Embase up to April 7, 2020. Outcomes of interest included clinical characteristics, molecular characteristic and survival analysis. HR (OR), odds ratios (OR) and 95% confidence interval (CI) were calculated to explore the prognostic value and relationship of clinical characteristics of Fusobacterium nucleatum in CRC. Results A total of 3626 CRC patients from 13 eligible studies were included. High levels of F. nucleatum were associated with worse prognosis, as such parameters as overall survival (OS) (hazard ratio [HR] = 1.40, 95% confidence interval [CI] 1.40 - 1.63, P less then 0.0001), disease-free survival (DFS) (HR = 1.71, 95% CI 1.29-2.26, P = 0.0002), and cancer-specific survival (OR= 1.93, 95% CI 1.42-2.62, P less then 0.0001). F. nucleatum levels were related with T3-T4 stage (OR = 2.20, 95% CI 1.66-2.91, P less then 0.00001), M1 stage (OR = 2.11, 95% CI 1.25-3.56, P = 0.005), poor tumor differentiation (OR = 1.83, 95% CI 1.11-3.03, P =0.02), microsatellite instability-high (OR = 2.53, 95% CI 1.53-4.20, P = 0.0003), and KRAS mutation (OR =1.27, 95% CI 1.00-1.61, P=0.05) showed. Conclusions High levels of F. nucleatum suggest a poor prognosis and are associated with tumor growth, distant metastasis, poor differentiation, MSI-high, and KRAS mutation in CRC patients.Background Small cell lung cancer (SCLC) represents about 13% of lung cancer cases, which is highly invasive and has a high mortality rate, with the 5-year overall survival (OS) rate being only 6.3%. Age at diagnosis of advanced SCLC is much older, but studies describing the ageing factor for distant metastasis patterns and prognosis of extensive-stage SCLC (ES-SCLC) are limited. Methods Using the Surveillance, Epidemiology, and End Results (SEER) registry, we identified 18,682 patients with ES-SCLC (9,089 women and 9,053 men) who had complete clinical information between 2008 and 2015. Patients were classified into three groups (older group ≥80 yrs, middle-aged group 60-79 yrs, and younger group ≤59 yrs). The role of different age at diagnosis of ES-SCLC (especially older group) in metastasis patterns was investigated, and OS and cancer-specific survival (CSS) of different age groups of metastatic ES-SCLC was assessed. Results The most metastasis of ES-SCLC patients in the three groups was multiorgan metastastases in elderly ES-SCLC patients. These results will be helpful for pre-treatment evaluation regarding the prognosis of ES-SCLC patients.In compliance with Article 43 of Regulation (EC) No 396/2005, the EFSA received from the European Commission a mandate to provide its reasoned opinion on the existing maximum residue levels (MRLs) for spinosad which might lead to consumers intake concerns on the basis of a new toxicological reference value established during the peer review and of the data currently available to EFSA. In order to identify the MRLs of potential concern that require a more detailed assessment, EFSA screened the existing MRLs for spinosad considering the new toxicological reference values and an acute risk could not be excluded for six commodities. Fall-back MRLs for all six commodities were proposed on the basis of the data received under the present assessment.The European Commission requested the EFSA Panel on Plant Health to prepare and deliver risk assessments for commodities listed in Commission Implementing Regulation (EU) 2018/2019 as 'High risk plants, plant products and other objects'. This Scientific Opinion covers the introduction risk for T. palmi posed by fruits of Momordica charantia L. imported from Thailand, taking into account the available scientific information, including the technical information provided by the Department of Agriculture of Thailand. The risk mitigation measures proposed in the technical dossier from Thailand were evaluated taking into account the possible limiting factors. An expert judgement is given on the likelihood of pest freedom taking into consideration the potential pest pressure in the field, the risk mitigation measures acting on the pest in the field and in the packing house, including uncertainties associated with the assessment.
