Different methods for determining the thermal inactivation kinetics of microorganisms can result in discrepancies in thermal resistance values. In this study, thermal resistance of Salmonella in whole milk powder was determined with three methods thermal death time (TDT) disk in water bath, pouches in water bath, and the TDT Sandwich system. Samples from three production lots of whole milk powder were inoculated with a five-strain Salmonella cocktail and equilibrated to a water activity of 0.20. The samples were then subjected to three isothermal treatments at 75, 80, or 85°C. Samples were removed at six time points and cultures were enumerated for survivors. The inactivation data were fitted to two consolidated models two primary models (log linear and Weibull) and one secondary model (Bigelow). Normality testing indicated that all the model parameters were normally distributed. None of the model parameters for both consolidated models were significantly different (α = 0.05). The amount of inactivation during the come-up time was also not significantly different among the methods (α = 0.05). However, the TDT Sandwich resulted in less inactivation during the come-up time and overall less variation in model parameters. The survivor data from all three methods were combined and fitted to both consolidated models. The Weibull had a lower root mean square error and a better fit, according to the corrected Akaike's information criterion. The three thermal treatment methods produced results that were not significantly different; thus, the methods are interchangeable, at least for Salmonella in whole milk powder. Comparisons with more methods, other microorganisms, and larger varieties of food products using the same framework presented in this study could provide guidance for standardizing thermal inactivation kinetics studies for microorganisms in foods. In this work, the goal was to determine the efficacy of MicroTally-based sampling in scenarios commonly encountered in the commercial beef processing industry, but outside of the parameters evaluated during the initial proof-of-concept work. The data were derived from 1,650 matched samples collected from 540 individual combo bins at six commercial beef processing plants, comparing MicroTally-based sampling (continuous and manual sampling devices [CSD and MSD]) to N60 Excision and/or N60 Plus methods. https://www.selleckchem.com/products/npd4928.html Mounting a 61-cm CSD cartridge to a 30-cm-wide conveyor provided sampling that is equivalent to N60 Excision and N60 Plus methods. Mounting a CSD to a chute instead of a conveyor was equivalent to the N60 Plus sampling method. The CSD was shown to be effective for sampling when used in conjunction with a "swinging arm trim diverter" and receiving product in batch mode as opposed to continuous flow. MSD sampling of oval combo bins with trim surface area (≈0.93 m2 [≈1,439 in2]) less than 1 m2 (1,600 in2) was shown to be equivalent to the N60 Plus sample collection method. Peracetic acid applied at the end of the trim conveyor did not negatively impact pathogen index target detection of the CSD even if the samples were shipped overnight before analysis. Pathogen index targets were demonstrated to be useful tools for validating methods designed to measure pathogen prevalence. The data presented herein support equivalency criteria of within 0.5 log CFU per sample for indicator organism counts. These data collectively support various alternative applications of MicroTally-based trim sampling and the application and interpretation of alternative methods for pathogen detection. Anti-mullerian hormone (AMH) is a promising biomarker for ovarian reserve. In this study, we assessed AMH before and one year after initiation of adjuvant chemotherapy on NSABP/NRG Oncology B-47 in female participants aged 42 years and younger (with median age 39 years). At baseline, median AMH was 1.2 ng/mL; 13 (4.7%) values were less then 0.1ng/mL (the threshold for detectable levels, in the perimenopause/menopause range), and 57 values (20.6%) were less than 0.5 ng/mL. At 1 year, 215 (77.6%) were less than 0.1ng/mL, and 264 (95.3%) were less than 0.5 ng/mL. Post-chemotherapy menses were reported by 46.2% of participants. Multivariable logistic regression found that the odds of having post-chemotherapy menses increased with younger age, higher BMI, and higher pre-chemotherapy AMH, but not by trastuzumab administration or by the choice of chemotherapy (doxorubicin-cyclophosphamide followed by paclitaxel vs. docetaxel-cyclophosphamide). We conclude that higher pre-chemotherapy AMH predicts a lower risk of chemotherapy-induced amenorrhea, and that AMH one year after chemotherapy initiation is not informative in this setting because it is likely to be very low. The occurrence, concentrations, and variables associated with tetracycline, polyether ionophore, and anthelmintic residues in the livers of chickens sold in the informal market in South Africa were determined. Ultrahigh-performance liquid chromatography-tandem mass spectrometry was used to simultaneously analyze for four tetracyclines, five polyether ionophores, and six anthelmintic residues. The study determined the presence of residues in liver samples at both the limit of quantifications (LOQ) and concentrations over the maximum residue limit (MRL), i.e., noncompliant. Doxycycline (tetracycline group) was detected in 24 (24.5%) of 98 chicken livers, and 15 (15.3%) of the 98 were noncompliant. Mean ± standard deviation (SD) concentrations of 919.04 ± 1,081.30 ppb (LOQ) and 1,410.57 ± 108.89 ppb (MRL) were obtained. Maduramicin was detected in 27 (27.6%) of 98 chicken livers, and 19 (19.4%) of 98 were noncompliant. The mean ± SD for LOQ was 117.96 ± 84.56 ppb, and MRL was 153.21 ± 76.29 ppb. The concentrations of residues of doxycycline and maduramicin in chicken livers varied significantly across townships. Lasalocid was found in 31 (31.6%) of 98 samples, of which 5 (5.1%) had concentrations above the MRL. The mean ± SD concentration of lasalocid was 62.90 ± 170.84 ppb for samples in which lasalocid was quantified and 310.16 ± 356.68 ppb for noncompliant samples. Detectable concentrations of praziquantel, closantel, and rafoxanide (anthelmintics) residues were found in 3 (3.1%), 1 (1.0%), and 2 (2.0%) of 98 chicken livers, respectively. The presence of residues of three classes of veterinary drugs in chicken livers poses food safety implications to consumers and indicates a need for enhanced regulatory enforcement in controlling these drugs in South Africa.
