These patients are at risk due to the reduced number of surgical aortic valve replacements and competition for intensive care facilities during the COVID-19 pandemic. This case series suggests that the minimally invasive transcatheter aortic valve implantation is safe to continue during the COVID-19 pandemic with adjustments to the patient pathway to minimize hospital stay and to reduce patient and staff exposure. This helps to reduce the delay of definitive treatment for patients with severe aortic stenosis.This article describes the development of a suitable Gel Permeation Chromatography method for cellulose nitrate plasticised with camphor (celluloid) found in cultural heritage. Current sample preparation and dissolution methods, apart from focusing on native, nonderivatised cellulose, require long preparation times, and often employ solvents that induce degradation. This study aims to develop a systematic method for sample preparation of cellulose nitrate that uses the least sample amount possible, is nondegrading, and can be applied on differently aged samples. This is investigated through identification of a suitable solvent system and a statistically designed experiment testing the critical variables affecting the analysis, namely sample condition, sample, and salt concentration (lithium chloride) in N,N-dimethylacetamide. The use of 0.1% sample was inadequate for analysis because it did not fully dissolve in any salt concentration, while the 0.3% negatively impacted the analysis with its high molecular weight distributions. The 0.2% cellulose nitrate in a solution of 0.5% lithium chloride in N,N-dimethylacetamide offered the most consistent and repeatable molecular weight data. This method miniaturised the sample as much as possible and is suitable for museum objects in various ageing conditions.Particulate matter in the air seriously affects human health and has been a hot topic of discussion. Because of the coronavirus disease 2019 (COVID-19) lockdown in cities in China, sources of particulate matter, including gasoline-burning vehicles, dust-producing building sites, and coal-fired factories, almost all ceased at the end of January 2020. It was not until early April that outdoor activities recovered. Ten cities were selected as observation sites during the period from 19 December 2019 to 30 April 2020, covering the periods of preclosure, closure, and gradual resumption. A total of 11 720 groups of data were obtained, and 4 indicators were used to assess the characteristics of the particle pollution in the period. The quality of the atmospheric environment was visibly influenced by human activities in those 5 mo. The concentrations of particulate matter with particle sizes below 10 µm (PM10) decreased slightly in February and March and then began to increase slowly after April with the gradual recovery of production. The concentrations of particulate matter with particle sizes below 2.5 µm (PM2.5) decreased greatly in most regions, especially in northern cities, during closure and maintained a relatively stable level in the following 3 mo. The trends of PM10 and PM2.5 indicated that the reduced human activities during the COVID-19 lockdown decreased the concentrations of particulate matter in the air, and the difference between the PM10 and PM2.5 trends might be due to the different sources of the 2 particles and their different aerodynamics. However, during closure, the particulate matter pollution in the cities remained at a high level, which indicated that some ignored factors other than outdoor production activities, automobile exhaust, and construction site dust might have contributed greatly to the PM10 and PM2.5 concentrations, and the tracing of the particulate matter should be given further attention in environmental management. Integr Environ Assess Manag 2021;001-11. © 2021 SETAC.Despite tremendous success of chimeric antigen receptor (CAR) T cell therapy in clinical oncology, the dose-exposure-response relationship of CAR-T cells in patients is poorly understood. Moreover, the key drug-specific and system-specific determinants leading to favorable clinical outcomes are also unknown. Here we have developed a multiscale mechanistic pharmacokinetic (PK)-pharmacodynamic (PD) model for anti-B-cell maturation antigen (BCMA) CAR-T cell therapy (bb2121) to characterize (i) in vitro target cell killing in multiple BCMA expressing tumor cell lines at varying effector to target cell ratios, (ii) preclinical in vivo tumor growth inhibition and blood CAR-T cell expansion in xenograft mice, and (iii) clinical PK and PD biomarkers in patients with multiple myeloma. Our translational PK-PD relationship was able to effectively describe the commonly observed multiphasic CAR-T cell PK profile in the clinic, consisting of the rapid distribution, expansion, contraction, and persistent phases, and accounted for the categorical individual responses in multiple myeloma to effectively calculate progression-free survival rates. Preclinical and clinical data analysis revealed comparable parameter estimates pertaining to CAR-T cell functionality and suggested that patient baseline tumor burden could be more sensitive than dose levels toward overall extent of exposure after CAR-T cell infusion. Virtual patient simulations also suggested a very steep dose-exposure-response relationship with CAR-T cell therapy and indicated the presence of a "threshold" dose, beyond which a flat dose-response curve could be observed. Our simulations were concordant with multiple clinical observations discussed in this article. Moving forward, this framework could be leveraged a priori to explore multiple infusions and support the preclinical/clinical development of future CAR-T cell therapies.
