Based on field efforts in Lake IJssel (the Netherlands) from 2000 to 2010, dynamics of 15 priority PAHs in suspended particulate matter (SPM) and in seawater were determined. This study examined time-varying changes in PAH concentration and composition, apportioned potential anthropogenic sources, and quantified their linkages to hydrological factors. The sum of individual PAHs (defined as ∑15 PAHs) varied widely throughout the study period in Lake IJssel, ranging from 0.76 to 9.66 mg/kg in SPM and from 0.014 to 0.136 μg/L in seawater. High molecular weight PAHs were the most abundant PAH compounds, responsible for 73%-97%. Seasonal variation in ∑15 PAHs level was distinguished, peaked in late winter and early spring, and reached a minimum in summer. Over site, the heavy contamination was found at the river mouth (e.g., Genemuiden). The driving factors of SPM and water temperature controlled the fate of PAHs. Principal component analysis and diagnostic ratios suggested that PAHs may have been derived from different input sources through various transport pathways, for example, coal combustion source and petroleum source. The findings are useful for filling the knowledge gap of PAH dynamics in Lake IJssel and Wadden Sea, which exerts significant influences on lake's environment. METHOD AND RESULTS Protective antigen (PA) and BA3338, a surface layer homology (SLH) domain possessing protein were cloned, expressed in heterologous system and purified by IMAC. Recombinant PA and BA3338 with alum were administered in mouse alone or in combination. The humoral and cell-mediated immune response was measured by ELISA and vaccinated animals were challenged with B. anthracis spores via intraperitoneal route. The circulating IgG antibody titre of anti-PA and anti-BA3338 was found significantly high in the first and second booster sera. A significant enhanced level of IL-4, IFN-γ and IL-12 was observed in antigens stimulated supernatant of splenocytes of PA + BA3338 vaccinated animals. A combination of PA and BA3338 provided 80% protection against 20 LD50 lethal dose of B. anthracis spores. CONCLUSION Both antigens induced admirable humoral and cellular immune response as well as protective efficacy against B. anthracis spores. SIGNIFICANCE AND IMPACT OF THE STUDY This study has been evaluated for the first time using BA3338 as a vaccine candidate alone or in combination with well-known anthrax vaccine candidate PA. The findings of this study demonstrated that BA3338 could be a co-vaccine candidate for development of dual subunit vaccine against anthrax. © 2020 The Society for Applied Microbiology.AIMS This research was conducted to investigate the biocatalytic remediation of xenobiotics polluted seawater using two biocatalysts; whole bacterial cells of facultative aerobic halotolerant Corynebacterium variabilis Sh42 and its extracted crude enzymes. METHODS AND RESULTS One-Factor-at-A-Time technique and statistical analysis were applied to study the effect of initial substrate concentrations, pH, temperature, and initial biocatalyst concentrations on the batch biocatalytic degradation of three xenobiotic pollutants (2-hydroxybiphenyl (2-HBP), catechol and benzoic acid) in artificial seawater (salinity 3·1%). HPLC and gas-chromatography mass spectroscopy analyses were utilized to illustrate the quantitative removal of the studied aromatic xenobiotic pollutants and their catabolic pathway. The results revealed that the microbial and enzymatic cultures followed substrate inhibition kinetics. Yano and Koga's equation showed the best fit for the biokinetic degradation rates of 2-HBP and benzoic acid, whereah42 is tolerable for toxic xenobiotics and different physicochemical environmental parameters. Thus, it can be recommended as an effective biocatalyst for biocatalytic remediation of xenobiotics polluted seawater. © 2020 The Society for Applied Microbiology.Congenital hepatic fibrosis (CHF) often accompanies autosomal recessive polycystic kidney disease (ARPKD), which stems from a PKHD1 gene mutation. The aim of this study was to clarify the prognosis of children with CHF who received living donor liver transplantation (LDLT) from donors who might be heterozygous carriers of a hepatorenal fibrocystic disease. Fourteen children with CHF who underwent LDLT at our center were enrolled. Eight and two patients had ARPKD and nephronophthisis, respectively. Eight of the donors were the recipients' fathers, and six donors were their mothers. We examined the histological and radiological findings of the donor livers and complications in the recipients following the liver transplantation. Seven of the donor livers presented morphological abnormalities of the bile ducts. Abdominal computed tomography revealed liver cysts in eight donors. One recipient underwent re-LT for graft failure due to rejection. Three patients presented with rejection, and one presented with sepsis. The overall survival rate was 100% and the original graft survival rate was 93%. In conclusion, the prognosis of recipients who received a LDLT from their parents for CHF was excellent. However, the morphology of half the donor livers was abnormal. Careful follow-up is needed to ensure long-term graft survival. © 2020 Japanese Society of Pathology and John Wiley & Sons Australia, Ltd.This research aimed to obtain the depth dependence of polymerization contraction and microporosity from irradiated dental resin cements by X-ray computed microtomography (μCT). Samples (n = 5) of commercial Relyx U200 (RU) and AllCem Core (AC) dual-cure resin cements were injected in a cylindrical Teflon sampler (25 mm3 ) and separated according to polymerization mechanism self-cured (not irradiated) and dual-cured (irradiated from the top surface with a LED device). The cement's volume was scanned with the μCT scanning conditions kept constant. To assess the depth dependence of polymerization contraction, it was measured the displacement of the cement mass from the sample holder at 30 vertical cuts (0.1 mm distant). To probe the microporosity, the percentage of area with presence of porosity by slice was obtained. All data were statistically treated. It was observed a positive linear correlation between depth and polymerization contraction in the irradiated groups. In the other hand, the concentration of micropores decreased with increasing depth.
