Puerto Rican adults have higher odds of peripheral artery disease (PAD) compared with Mexican Americans. Limited studies have examined relationships between clinical risk assessment scores and ABI measures in this population. Using 2004-2015 data from the Boston Puerto Rican Health Study (BPRHS) (n = 370-583), cross-sectional, 5-y change, and patterns of change in Framingham Risk Score (FRS) and allostatic load (AL) with ankle brachial index (ABI) at 5-y follow-up were assessed among Puerto Rican adults (45-75 y). FRS and AL were calculated at baseline, 2-y and 5-y follow-up. Multivariable linear regression models were used to examine cross-sectional and 5-y changes in FRS and AL with ABI at 5-y. Latent growth mixture modeling identified trajectories of FRS and AL over 5-y, and multivariable linear regression models were used to test associations between trajectory groups at 5-y. Greater FRS at 5-y and increases in FRS from baseline were associated with lower ABI at 5-y (β = -0.149, P = 0.010; β = -0.171, P = 0.038, respectively). AL was not associated with ABI in cross-sectional or change analyses. Participants in low-ascending (vs. no change) FRS trajectory, and participants in moderate-ascending (vs. low-ascending) AL trajectory, had lower 5-y ABI (β = -0.025, P = 0.044; β = -0.016, P = 0.023, respectively). FRS was a better overall predictor of ABI, compared with AL. Puerto Rican adults, an understudied population with higher FRS over 5 years, may benefit from intensive risk factor modification to reduce risk of PAD. Additional research examining relationships between FRS and AL and development of PAD is warranted.The purpose of this study was to evaluate the effects of an industrial passive assisted exoskeleton (IPAE) with simulated lifting tasks on muscle activity, oxygen consumption, perceived level of exertion, local perceived pressure, and systemic usability. Eight workers were required to complete two lifting tasks with and without the IPAE, that were single lifting tasks (repeated 5 times) and 15 min repeated lifting tasks respectively. Both of the tasks required subjects to remove a toolbox from the ground to the waist height. The test results showed that IPAE significantly reduced the muscle activity of the lumbar erector spinae, thoracic erector spinae, middle deltoid and labrum-biceps muscles; the reduction effect during the 15 min lifting task was reached 21%, 12%, 32% and 38% respectively. However, there is still a concern for the obvious contact pressure.During oropharyngeal candidiasis (OPC), Candida albicans invades and damages oral epithelial cells, which respond by producing proinflammatory mediators that recruit phagocytes to foci of infection. The ephrin type-A receptor 2 (EphA2) detects β-glucan and plays a central role in stimulating epithelial cells to release proinflammatory mediators during OPC. The epidermal growth factor receptor (EGFR) also interacts with C. albicans and is known to be activated by the Als3 adhesin/invasin and the candidalysin pore-forming toxin. Here, we investigated the interactions among EphA2, EGFR, Als3 and candidalysin during OPC. We found that EGFR and EphA2 constitutively associate with each other as part of a heteromeric physical complex and are mutually dependent for C. albicans-induced activation. Als3-mediated endocytosis of a C. albicans hypha leads to the formation of an endocytic vacuole where candidalysin accumulates at high concentration. Thus, Als3 potentiates targeting of candidalysin, and both Als3 and candidalysin are required for C. albicans to cause maximal damage to oral epithelial cells, sustain activation of EphA2 and EGFR, and stimulate pro-inflammatory cytokine and chemokine secretion. In the mouse model of OPC, C. albicans-induced production of CXCL1/KC and CCL20 is dependent on the presence of candidalysin and EGFR, but independent of Als3. The production of IL-1α and IL-17A also requires candidalysin but is independent of Als3 and EGFR. The production of TNFα requires Als1, Als3, and candidalysin. Collectively, these results delineate the complex interplay among host cell receptors EphA2 and EGFR and C. albicans virulence factors Als1, Als3 and candidalysin during the induction of OPC and the resulting oral inflammatory response.While the international pet trade and habitat destruction have been extensively discussed as major threats to the survival of the pancake tortoise (Malacochersus tornieri), the impact of climate change on the species remains unknown. In this study, we used species distribution modelling to predict the current and future distribution of pancake tortoises in Zambezian and Somalian biogeographical regions. We used 224 pancake tortoise occurrences obtained from Tanzania, Kenya and Zambia to estimate suitable and stable areas for the pancake tortoise in all countries present in these regions. We also used a protected area network to assess how many of the suitable and stable areas are protected for the conservation of this critically endangered species. Our model predicted the expansion of climatically suitable habitats for pancake tortoises from four countries and a total area of 90,668.75 km2 to ten countries in the future and an area of 343,459.60-401,179.70 km2. The model also showed that a more significant area of climatically suitable habitat for the species lies outside of the wildlife protected areas. Based on our results, we can predict that pancake tortoises may not suffer from habitat constriction. However, the species will continue to be at risk from the international pet trade, as most of the identified suitable habitats remain outside of protected areas. We suggest that efforts to conserve the pancake tortoise should not only focus on protected areas but also areas that are unprotected, as these comprise a large proportion of the suitable and stable habitats available following predicted future climate change.
