In this review, the most important neuropathological changes found in the cerebella of sheep affected by classical natural scrapie are discussed. This disease is the oldest known of a group of unconventional "infections" caused by toxic prions of different origins. Scrapie is currently considered a "transmissible spongiform encephalopathy" (due to its neuropathological characteristics and its transmission), which is the paradigm of prion pathologies as well as many encephalopathies (prion-like) that present aberrant deposits of insoluble protein with neurotoxic effects due to errors in their catabolization ("misfolding protein diseases"). However, the transferability to real-world scenarios cannot be granted due to limitations on data and evaluation methods. On the example of football shot and pass detection against a null class we explore the influence of those factors for real-world event classification in field sports. For this purpose we compare the performance of an established Support Vector Machine (SVM) for laboratory settings from literature to the performance in three evaluation scenarios gradually evolving from laboratory settings to real-world scenarios. In addition, three different types of neural networks, namely a convolutional neural net (CNN), a long short term memory net (LSTM) and a convolutional LSTM (convLSTM) are compared. Results indicate that the SVM is not able to reliably solve the investigated three-class problem. In contrast, all deep learning models reach high classification scores showing the general feasibility of event detection in real-world sports scenarios using deep learning. The maximum performance with a weighted f1-score of 0.93 was reported by the CNN. The study provides valuable insights for sports assessment under practically relevant conditions. In particular, it shows that (1) the discriminative power of established features needs to be reevaluated when real-world conditions are assessed, (2) the selection of an appropriate dataset and evaluation method are both required to evaluate real-world applicability and (3) deep learning-based methods yield promising results for real-world HAR in sports despite high variations in the execution of activities.New substituted azomethines of benzanthrone with heterocyclic substituents were synthesized by condensation reaction of 3-aminobenzo[de]anthracen-7-one with appropriate aromatic aldehydes. The resulting imines were reduced with sodium borohydride to the corresponding amines, the luminescence of which is more pronounced in comparison with the initial azomethines. The novel benzanthrone derivatives were characterized by NMR, IR, MS, UV/Vis, and fluorescence spectroscopy. The structure of three dyes was studied by the X-ray single crystal structure analysis. The solvent effect on photophysical behaviors of synthesized imines and amines was investigated. The obtained compounds absorb at 420-525 nm, have relatively large Stokes shifts (up to 150 nm in ethanol), and emit at 500-660 nm. The results testify that emission of the studied compounds is sensitive to the solvent polarity, exhibiting negative fluorosolvatochromism for the synthesized azomethines and positive fluorosolvatochromism for the obtained amines. Particularly when imaging inflammation, the technology offers advantages over current clinical methods to diagnose, stage, and monitor physiological and pathophysiological processes. This review discusses the clinical perspective of using OAI in the context of imaging inflammation as well as in current and emerging translational applications.Endothelium has a rich vesicular network that allows the exchange of macromolecules between blood and parenchymal cells. This feature of endothelial cells, along with their polarized secretory machinery, makes them the second major contributor, after platelets, to the particulate secretome in circulation. Extracellular vesicles (EVs) produced by the endothelial cells mirror the remarkable molecular heterogeneity of their parent cells. Cargo molecules carried by EVs were shown to contribute to the physiological functions of endothelium and may support the plasticity and adaptation of endothelial cells in a paracrine manner. Endothelium-derived vesicles can also contribute to the pathogenesis of cardiovascular disease or can serve as prognostic or diagnostic biomarkers. Finally, endothelium-derived EVs can be used as therapeutic tools to target endothelium for drug delivery or target stromal cells via the endothelial cells. In this review we revisit the recent evidence on the heterogeneity and plasticity of endothelial cells and their EVs. We discuss the role of endothelial EVs in the maintenance of vascular homeostasis along with their contributions to endothelial adaptation and dysfunction. Finally, we evaluate the potential of endothelial EVs as disease biomarkers and their leverage as therapeutic tools.
