This site is defined by an N-terminal histidine and a second internal histidine side chain in a configuration called the histidine brace. To understand better the determinants of reactivity, the biochemical and structural properties of a well-described cellulose-specific LPMO from Thermoascus aurantiacus (TaAA9A) is compared with that of CopC from Pseudomonas fluorescens (PfCopC) and with the LPMO-like protein Bim1 from Cryptococcus neoformans. PfCopC is not reduced by ascorbate but is a very strong Cu(II) chelator due to residues that interacts with the N-terminus. This first biochemical characterization of Bim1 shows that it is not redox active, but very sensitive to H2O2, which accelerates the release of Cu ions from the protein. TaAA9A oxidizes ascorbate at a rate similar to free copper but through a mechanism that produce fewer reactive oxygen species. These three biologically relevant examples emphasize the diversity in how the proteinaceous environment control reactivity of Cu with O2.Domestic dogs are frequently encountered in Indigenous archaeological sites on the Northwest Coast of North America. Although dogs depended on human communities for care and provisioning, archaeologists lack information about the specific foods dogs consumed. Previous research has used stable isotope analysis of dog diets for insight into human subsistence ('canine surrogacy' model) and identified considerable use of marine resources. Here, we use zooarchaeological data to develop and apply a Bayesian mixing model (MixSIAR) to estimate dietary composition from 14 domestic dogs and 13 potential prey taxa from four archaeological sites (2,900-300 BP) in Tseshaht First Nation territory on western Vancouver Island, British Columbia, Canada. Two candidate models that best match zooarchaeological data indicate dogs predominantly consumed salmon and forage fish (35-65%), followed by nearshore fish (4-40%), and marine mammals (2-30%). We compared these isotopic data to dogs across the Northwest Coast, which indicated a pronounced marine diet for Tseshaht dogs and, presumably, their human providers. The prognosis of CLL-associated PNP is dismal due to its refractory course and secondary infection and no standard treatment was recommended. We retrospectively reported six CLL with PNP cases from 842 cases of CLL including diagnosis, treatment and prognosis. The median time between the initial of CLL to PNP was 36 months while the median overall survival from the diagnosis of PNP was 26 months. And three cases died of lung infection while 5 developed pulmonary symptoms. And 5 cases received fludarabine-based chemotherapy before developing PNP, which suggesting fludarabine was one of potential causes of PNP. For the treatment, five patients were rescued by combined regimens including rituximab, methylprednisolone, immunoglobulin, fresh frozen plasma and the last received ibrutinib combined with short-term prednisone. Fludarabine-based regimen may be one of the potential causes of PNP. The combined regimen might shed a new light, while ibrutinib is a promising drug for CLL with PNP, but needs much more evidence. PNP should be carefully treated to guide early diagnosis and intervention for a better prognosis.Palaeoenvironmental reconstructions of the interior of South Africa show a wetter environment than today and a non-analogous vegetation structure in the Early Pleistocene. This includes the presence of grasses following both C3 and C4 photosynthetic pathways, whereas C3 grasses decline after the mid-Pleistocene transition (MPT, c. 1.2-0.8 Ma). However, the local terrestrial proxy record cannot distinguish between the potential drivers of these vegetation changes. In this study we show that low glacial CO2 levels, similar to those at the MPT, lead to the local decline of C3 grasses under conditions of decreased water availability, using a vegetation model (LPX) driven by Atmosphere-Ocean coupled General Climate Model climate reconstructions. We modelled vegetation for glacial climates under different levels of CO2 and fire regimes and find evidence that a combination of low CO2 and changed seasonality is driving the changes in grass cover, whereas fire has little influence on the ratio of C3C4 grasses. Our results suggest the prevalence of a less vegetated landscape with limited, seasonal water availability, which could potentially explain the much sparser mid-Pleistocene archaeological record in the southern Kalahari.
