In the veterinary profession, the body condition score (BCS) plays an important role in the assessment of patients. It is a subjective, tactile method of evaluating body fat and muscle mass and is used in numerous species. Recognizing obesity (or the contrary, emaciation) is important for veterinarians treating reptiles and could be facilitated by a BCS. An existing form of body condition assessment already used is the body condition index (BCI), where the residuals from a regression of body mass on body length are calculated. Therefore, the goal of this study was to provide practitioners with a BCS system for corn snakes (Pantherophis guttatus) and to test it against the BCI. A total of 22 corn snakes (Pantherophis guttatus), stationed at the "Auffangstation für Reptilien" in Munich (reptile rescue centre, RRC), were subject of this study. Each had the following measurements taken body weight (BW), snout-tail tip length (STL), snout-vent length (SVL) and circumference in the middle (C). Manual palpation of spine, area between vertebral spinous and transverse process, ribs and neck of each snake was performed by three veterinarians and assigned to specific scores by each examiner. A BCS (mean of examiners' scores) was given to each snake according to manual palpation. The BCS system was chosen to be out of 5 in 0.5-point steps with 2.5 considered as ideal BCS. In the studied snakes, the BCS ranged from 1.5 to 3.5, with a median of 2.5. The median BW was 309 g (75-967 g), the median STL was 123 cm (79-153 cm), the median SVL was 104 cm (73-133 cm) and the median C was 7.5 cm (4.3-11 cm). BCS and BCI were positively correlated. A BCS includes a manual palpation of the animal and thus gives the examiner additional information to the objectively measured/calculated index. © 2020 Blackwell Verlag GmbH.While teaching, the Institute of Animal Nutrition of the University of Zurich (IANZ) noticed that veterinary students, veterinarians and pet owners have significant difficulties in reading and understanding pet food labels (PFL). The authors hypothesized that if PFL are easy to read and understand, 90% of those dog owners (n = 43) polled at a dog exhibition are able to answer a questionnaire after reading an exemplary PFL modified from existing products. About 30.2% of the participants were able to compare the protein content of a canned diet versus a dry diet. About 34.9% were able to estimate and compare the carbohydrate content of two dry diets by reading the composition. About 32.6% identified which of the two dry diets was potentially a complete diet by reading the listed ingredients. The results show that a majority of the participants was overwhelmed with the task of reading and understanding the exemplary PFL. An improvement of public education on the basics of product labelling or additional simplified information on the labels might be helpful. © 2020 Blackwell Verlag GmbH.AIMS To examine whether World Health Organization (WHO) risk-level reductions in drinking were achievable, associated with improved functioning and maintained over time among patients at varying initial alcohol dependence severity levels. Design and setting Secondary data analysis of multi-site randomized clinical trials the US Combined Pharmacotherapies and Behavioral Interventions for Alcohol Dependence (COMBINE) study and the UK Alcohol Treatment Trial (UKATT). PARTICIPANTS Individuals with alcohol dependence enrolled in COMBINE (n = 1383; 68.8% male) and seeking treatment for alcohol problems in UKATT (n = 742; 74.1% male). Interventions Naltrexone, acamprosate or placebo, and combined behavioral intervention or medication management in COMBINE. Social behavior network therapy or motivational enhancement therapy in UKATT. MEASUREMENTS WHO risk-level reductions were assessed via the calendar method. Alcohol dependence was measured by the Alcohol Dependence Scale, the Leeds Dependence Questionnaire and the risk levels for alcohol consumption appear to be achievable, associated with better functioning and maintained over time in both the United States and the United Kingdom. © 2020 Society for the Study of Addiction.Kinesin family member 2C (KIF2C), a substantial mitotic regulator, has been verified to exert a malignant function in several cancers. However, its function in hepatocellular carcinoma (HCC) remains unclear. In this study, the expression profile of KIF2C in HCC was characterized through the dataset from the TCGA and clinical tissue microarrays containing 220 pairs of resected HCC tissues and adjacent nontumor tissues in our hospital. The results indicated that KIF2C was substantially higher expression in tumor tissues than adjacent nontumor tissues. High expression of KIF2C significantly correlated with large tumor (>5.0 cm) (P = .001) and implied a dismal postoperative overall survival (OS) (hazard ratio [HR] = 1.729; P = .002) in our cohort of patients. Gain and loss of function assays displayed that KIF2C promoted HCC cell proliferation, accelerated cell cycle progression, and impeded apoptosis. By bioinformatic tools and mechanistic investigation, we found that KIF2C interacted with various cell-cycle-related proteins and was significantly involved in growth-promoting pathways. KIF2C upregulated PCNA and CDC20 expression. Subsequently, we investigated the regulation of KIF2C by competing endogenous RNA and elucidated that has-miR-6715a-3p was directly bond to the 3'-untranslated region of KIF2C through dual luciferase assays, thereby inhibiting KIF2C expression. Furthermore, the long noncoding RNA GS1-358P8.4 was found to be a candidate of KIF2C for has-miR-6715a-3p binding. HCC patients with high lncRNA-GS1-358P8.4 expression had shorter OS and relapse-free survival compared to those with low expression, which was accordance with the KIF2C. Taken together, KIC2C aggravated HCC progression, it could serve as a prognostic indicator and confer a novel target for clinical treatment. © 2020 Wiley Periodicals, Inc.Aging skeletal muscle shows perturbations in metabolic functions. MicroRNAs have been shown to play a critical role in aging and metabolic functions of skeletal muscle. MicroRNA-34a (miR-34a) is implicated in the brain and cardiac aging, however, its role in aging muscle is unclear. https://www.selleckchem.com/products/mrtx0902.html We analyzed levels of miR-34a, ceramide kinase (CERK) and other insulin signaling molecules in skeletal muscle from old mice. In addition to in vivo model, levels of these molecules were also analyzed in myoblast derived from insulin resistant (IR) humans and C2C12 myoblasts overexpressing mir-34a. Our results show that miR-34a is elevated in the muscles of 2-year-old mice and in the myoblasts of IR humans. Overexpression of miR-34a in C2C12 myoblasts leads to alterations in the insulin signaling pathway, which were rescued by its antagonism. Our analyses revealed that miR-34a targets CERK resulting in ceramide accumulation, activation of PP2A and the pJNK pathway in muscle and C2C12 myoblasts. Also, myostatin (Mstn) levels were increased in 2-year-old mouse muscle and Mstn treatment upregulated miR-34a in C2C12 myoblasts.
