[18F]FBEM-Cys39-exendin-4, [18F]DPA-714, [18F]fallypride, and [18F]FDG were prepared and PET imaging acquisitions for 3×Tg-AD mice and wild-type (WT) mice were performed at 15, 30, and 60 min post-injection. Fifteen regions of interest (ROIs) were selected and %ID/g was calculated. The results showed that the uptake of [18F]FBEM-Cys39-exendin-4 in 10 ROIs of 3×Tg-AD mice at 60 min post-injection was significantly lower than that of WT mice (p less then 0.05). Besides, 3×Tg-AD mice showed significantly higher [18F]DPA-714 uptake in 7 ROIs and lower [18F]fallypride uptake in 4 ROIs compared to WT mice. [18F]FDG PET showed no significant differences in any ROIs between the two groups. A positive correlation between the uptake of [18F]fallypride and [18F]FBEM-Cys39-exendin-4 could be found in the whole brain. In summary, these results validated the feasibility of GLP-1R PET in AD and demonstrated the reduced GLP-1R and D2R expression as well as increased microglial activation caused by AD. Apelin, a specific endogenous ligand of the G protein-coupled receptor APJ, suppresses oxidative stress and apoptosis in vitro and in vivo. The current study explored whether Apelin protects against toxicity induced by the anticancer drug cisplatin in vitro, and the possible mechanisms that underlie this protective effect. The results showed that Apelin was expressed in the mouse auditory cell line HEI-OC1 and in cochlear hair cells (HCs) and was significantly downregulated by cisplatin, whereas pre-treatment with exogenous Apelin significantly reduced cisplatin-induced apoptosis, and thus protected HEI-OC1 cells and cochlear HCs from cisplatin-induced injury. Furthermore, Apelin reduced reactive oxygen species (ROS) generation, rescued mitochondrial membrane potential disruption, inhibited JNK signaling and attenuated the expression of pro-apoptotic factors in HEI-OC1 cells and in cochlear explants treated with cisplatin. V.Two new caged xanthones, wightiic acid (1) 16-O-methyl wightiic acid (2), along with eight known compounds, gaudichaudic acid E (3), isogaudichaudic acid E (4), ursolic acid (5) stigmasterol (6), lupeol (7), glutinol (8), lupenone (9) and stigmasteryl linoleate (10) were isolated from Garcinia wightii T. Anderson. The structures of the compounds were elucidated by spectroscopic means, including 1D and 2D NMR, HR-ESIMS, and the absolute configuration of the new compounds 1 and 2 were determined by analysis of ECD data. Anti-proliferation activities of the four caged xanthones (1-4) were evaluated by MTT assay on MCF-7 and SKBR-3 human breast cancer cells and A-375 human melanoma cells by MTT assay. Here, we analyzed role of DNA methylation under desiccation and salinity stresses in three (IR64, stress-sensitive; Nagina 22, drought-tolerant and Pokkali, salinity-tolerant) rice cultivars via bisulphite sequencing. Methylation in CG context within gene body and methylation in CHH context in distal promoter regions were positively correlated with gene expression. Hypomethylation in Nagina 22 and hypermethylation in Pokkali in response to desiccation and salinity stresses, respectively, were correlated with higher expression of few abiotic stress response related genes. Most of the differentially methylated and differentially expressed genes (DMR-DEGs) were cultivar-specific, suggesting an important role of DNA methylation in abiotic stress responses in rice in cultivar-specific manner. DMR-DEGs harboring differentially methylated cytosines due to DNA polymorphisms between the sensitive and tolerant cultivars in their promoter regions and/or coding regions were identified, suggesting the role of epialleles in abiotic stress responses.
